摘要
为解决家禽养殖业中大量羽毛废弃物及其污染问题,从环境中筛选生长速度快、产酶量高、应用潜力好的菌株进行研究.从土壤中分离得到一株高效降解羽毛的S-2菌株,对其菌落形态、生理生化特性及16S rRNA序列进行分析,并进一步分析在不同培养基中的羽毛降解率及最适产酶条件.结果发现该菌株与Bacillus aryabhattai QK2的同源度最高,初步认定该菌株为阿氏芽胞杆菌(Bacillus aryabhattai),命名为Bacillus aryabhattai S-2.菌S-2在发酵培养基中经过96 h培养,对羽毛降解率高达97.2%,在初始pH为7.0,接种量为2%,羽毛含量为0.5 g/50 mL,发酵培养时间为48 h,发酵温度为37℃的条件下产酶活性最大.表明该菌株对羽毛有较强的降解能力,为未来深入研究废弃羽毛的降解打下了基础.
In order to solve the problem of a large amount of feather waste and its pollution in the poultry breeding industry, screening strains with high growth rate, high enzyme production and good application potential from the environment has important research values. A bacterial strain with high efficiency in degrading poultry feather, strain S-2, was isolated from a soil sample. The strain was identified by morphological, physiological, 16 S rRNA and biochemical tests. Besides, the feather degradation rate and optimum enzyme production in different media were further analyzed. The results showed that this strain had the highest degree of homology with Bacillus aryabhattai QK2, so that it was named as Bacillus aryabhattai S-2. The degradation rate of B. aryabhatta S-2 reached 97.2% in the fermentation medium after 96 h of inoculation. The highest enzyme level was obtained at the initial pH of 7.0, the inoculation quantity of 2%, the fermentation temperature of 37 T and the incubation time of 48 h. The results indicated that the strain had a strong ability to degrade feather waste, which lays a foundation for studies of degradation of poultry feather in the future.
