盐胁迫下大麻SSR标记的分布特征分析 被引量:2

摘要 为探究盐胁迫对大麻遗传育种改良的影响,试验对盐胁迫条件下大麻转录组测序数据进行了SSR位点标记的挖掘,并对大麻转录组数据中SSR位点标记、SSR位点信息和序列分布特征等进行了分析。结果表明:共检测出84 483条Unigene序列,序列总长87.31 Mb。筛选出27 861个SSR位点,SSR标记的检出率为33.10%,即平均3.13 kb就会出现一个SSR位点,其中以单、二和三碱基重复为主,检出率分别为52.73%、23.26%和22.16%;重复基元主要为A/T、AG/CT、AT/AT和AAG/CTT,分别占52.57%、11.32%、10.25%和6.69%。通过Primer 3.0在线软件获得17 894条SSR引物序列,引物中SSR位点重复长度绝大多数集中在10~21 bp,占SSR重复总数的79.72%,其中最长重复长度为226 bp。该研究构建的盐胁迫下大麻SSR位点信息与分布特征分析,可为研究大麻分子标记辅助育种、EST-SSR标记的开发等提供信息资源。 In order to investigate the effect of salt stress on the improvement of hemp genetic breeding, this study was conducted to mine the sequencing data of hemp transcriptome under salt stress conditions for SSR site markers and analyzed the SSR site markers, SSR site information and sequence distribution characteristics of hemp transcriptome data. The study showed that a total of 84 483 Unigenes were detected, with a total of 87.31 Mb, 27 861 SSR sites were screened, and the detection rate of SSR markers was 33.10%, i.e. there was one SSR site in 3.13 kb on average. Mononucleotide, Dinucleotide, and Trinucleotide base repeats were predominant, with detection rates of 52.73%, 23.26% and 22.16%;the repeat primitives were mainly A/T, AG/CT, AT/AT and AAG/CTT, accounting for 52.57%, 11.32%, 10.25% and 6.69%, respectively. 17894 EST-SSR primer sequences were obtained through Primer 3.0 online software, and the vast majority of SSR site repeat lengths in primers were concentrated between 10 and 21 bp, accounting for 79.72% of the total SSR repeats, of which the longest repeat length was 226 bp. The analysis of the SSR locus information and distribution characteristics of hemp under salt stress constructed in this study can provide information resources for the research of molecular marker-assisted breeding of hemp and the development of EST-SSR markers.
