摘要 为探究朝天椒组织培养的最佳培养基配方,以MS、B_(5)、WPM为基础培养基成分,通过添加不同含量琼脂和蔗糖,在无菌条件下培养,分析不同培养基对朝天椒种子萌发及幼苗生长发育指标的影响。结果表明:9个配方处理中,WPM+9 g/L琼脂+30 g/L蔗糖处理种子发芽率100%,茎粗、株高、地下鲜重均最大,显著高于其他处理,WPM+琼脂(9 g/L)+蔗糖(30 g/L)为最佳培养基配方,生长的幼苗根冠比为0.3842,壮苗指数最高,组培苗质量最好,且经正交试验极差和方差分析三个因素对朝天椒幼苗生长影响的主次顺序为A(培养基)>C(蔗糖)>B(琼脂),其中A(培养基)的影响差异显著(P<0.05)。 In order to find out the optimal medium formula for tissue culture of pepper,used MS,B_(5) and WPM as basic medium components,and added into different contents of agar and sucrose to cultivate pepper under sterile conditions to study the effects of different media on seed germination and seedling growth of pepper.The results showed that among the nine formula treatments,WPM+Agar(9g/L)+sucrose(30g/L)was the optimal medium formula,on which the seed germination rate was 100%,the stem diameter,plant height and underground fresh weight were the highest,the root-shoot ratio was 0.3842,the strong seedling index was the highest,and the seedling quality was the best.The results of range and variance analysis of orthogonal test indicated that the influence order of the three factors on pepper seedling growth was A(medium)>C(Sucrose)>B(Agar),and the effect of A(medium)was significant(P<0.05).
