摘要
为研究半滑舌鳎Cynoglossu semilaevis Günther套肠症的病原,于2020年9月从天津市滨海新区某养殖车间采集患病鱼10尾(体长10~15 cm),利用寄生虫检查、病原菌分离、组织病理切片、人工感染试验、生理生化试验、16S rDNA基因序列分析和药物敏感性试验等方法对患病鱼及病原进行了研究。结果表明:患病鱼体表未发现大量寄生虫,脾脏中存在大量结节,肠黏膜组织和肠绒毛腐烂脱落,从肾脏中分离到的优势菌株ST-2020-2能引起健康半滑舌鳎发病,造成90%的死亡率;综合生理生化特征和16S rDNA基因序列分析结果,将其鉴定为美人鱼发光杆菌杀鱼亚种Photobacterium damselae subsp.piscicida;药敏试验显示,菌株ST-2020-2对链霉素、环丙沙星、氟苯尼考敏感,对四环素、卡那霉素、氨苄西林中度敏感,对新霉素、多黏菌素、复方新诺明等10种药物具有耐药性。研究表明,美人鱼发光杆菌杀鱼亚种是半滑舌鳎套肠症的病原,这为该病的防控提供了基础数据。
To investigate the pathogen responsible for the intussusception disease of half-smooth tongue sole Cynoglossus semilaevis Günther,parasite inspection,pathogen isolation,histopathological analysis,infection test,physiological and biochemical tests,and drug susceptibility tests were performed in 10 diseased fish with body length of 10-15 cm collected from a breeding plant in Tianjin in September 2020 by 16S rDNA gene sequence analysis.The results showed that there were no large numbers of parasites on the body surfaces of the diseased fish,whereas a large number of nodules were found in the spleens,with decayed and fallen away intestinal mucosal tissue and intestinal villi.The dominant bacterial strain ST-2020-2 was isolated from the kidney caused morbidity in healthy half-smooth tongue sole,with 90%mortality in the infection test.Integrating the physiological and biochemical characteristics and the 16S rDNA gene sequence analysis revealed that ST-2020-2 was identified as strain Photobacterium damselae subsp.piscicida and sensitive to streptomycin,ciprofloxacin and florfenicol,moderately sensitive to tetracycline,kanamycin and ampicillin,and resistant to 10 drugs,including neomycin,polymyxin B,and tobramycin.The findings indicated that Photobacterium mermaid was the pathogen of intestinal disease observed in the half-smooth tongue sole,and that provided essential data for the prevention and control of this disease.
