摘要
目的运用网络药理学的方法探讨松香酸治疗银屑病的分子作用机制。方法采用STRING数据库和Cytoscape软件绘制蛋白质-蛋白质相互作用网络。借助DAVID数据库对靶点进行基因本体分析和京都基因与基因组的百科全书通路分析,将核心靶点与松香酸进行分子对接,以验证松香酸与核心靶点的结合能力。结果筛选得到松香酸治疗银屑病的60个作用靶点,主要为肿瘤坏死因子、丝裂原活化蛋白激酶3、肿瘤蛋白53和过氧化物酶体增殖物激活受体(peroxisome proliferationactivated receptor,PPAR)γ。靶点主要涉及细胞锌离子结合、类固醇激素受体活性、维甲酸受体活性等生物过程。PPAR信号通路和花生四烯酸代谢信号通路为松香酸治疗银屑病的主要作用通路。结论松香酸治疗银屑病体现了多靶点、多途径的作用特点,该研究为深入阐释松香酸治疗银屑病作用机制提供了科学依据。
Objective To explore the molecular mechanism of abietic acid in the trearment of psoriasis by using a network pharmacology.Methods Proteinprotein interaction networks were mapped using STRING database and Cytoscape software.Gene Ontology analysis and Kyoto Encyclopedia of Genes and Genomes analysis of targets were performed on the target with the help of DAVID database.Molecular docking was performed between the core target and abietic acid to verify the binding ability of abietic acid to the core target.Results Sixty targets for the treatment of psoriasis with abietic acid were screened,mainly tumor necrosis factor(TNF),mitogen-activated protein kinase 3(MAPK3),tumor protein 53(TP53)and peroxisome proliferation-activated receptorγ(PPAR).The targets mainly involved in cell zinc ion binding,steroid hormone receptor activity,retinoic acid receptor activity and other biological processes.Peroxisome proliferation-activated receptor(PPAR)signaling pathway and arachidonic acid metabolism signaling pathway are the main action pathways of abietic acid in the treatment of psoriasis.Conclusion Abietic acid treatment of psoriasis reflects the characteristics of multitarget and multi-pathway.This study provides a scientific basis for further explaining the mechanism of abietic acid in the treatment of psoriasis.
