摘要
目的:观察藤黄健骨片对膝关节骨关节炎(KOA)模型大鼠急性发作期骨代谢、行为学及核因子-κB(NF-κB)、细胞间黏附分子-1(ICAM-1)表达的影响。方法:大鼠膝关节腔注射木瓜蛋白酶制备KOA模型。实验分为正常组、模型组、阳性药组和藤黄健骨片低、中、高剂量组,每组10只。正常组与模型组生理盐水灌胃,阳性组给予秋水仙碱灌胃,其余大鼠均予以不同剂量的藤黄健骨片灌胃,连续灌胃14 d。观察各组大鼠骨代谢、行为学变化,RT-PCR、Western blot检测NF-κB mRNA及ICAM-1 mRNA表达,ELISA检测血清炎症因子(IL-6、IL-1β、CRP)水平。结果:蛋白聚糖水平藤黄健骨片各剂量组、阳性组均高于模型组,阳性组高于低剂量组和中剂量组(均P<0.05)。各剂量组行为学异常占比均低于模型组(均P<0.05)。各剂量组、阳性组NF-κB mRNA、ICAM-1 mRNA及蛋白表达均低于模型组,阳性组低于低剂量组和中剂量组(均P<0.05)。各剂量组、阳性组血清IL-6、IL-1β、CRP水平均低于模型组,阳性组低于低剂量组和中剂量组(均P<0.05)。结论:藤黄健骨片可有效改善KOA急性发作期模型大鼠骨代谢、行为学变化,其主要作用机制可能和下调NF-κB、ICAM-1表达以及炎症因子水平有关。
Objective:To investigate the effects of Tenghuang Jiangu Tablet on bone metabolism,behavior,expressions of nuclear factor-κB(NF-κB)and intercellular adhesion molecule-1(ICAM-1)in KOA model rats at acute exacerbation stage.Methods:The KOA model was established by injecting Papain into the knee joint,experiment was divided into normal,model,positive and Tenghuang Jiangu Tablet low,middle,high dose groups.With each 10 rats.The normal and model groups were given saline by gavage,the positive group was given Colchicine by gavage,the other rats were given different doses of Tenghuang Jiangu tablets by gavage.The changes of bone metabolism,behavior were observed,expressions of NF-κB,ICAM-1 mRNA and protein were determined by RT-PCR and Western blot,and serum inflammatory factors(IL-6,IL-1β,CRP)were determined by ELISA.Results:The proteoglycan level of each administration group were higher than that of model group(all P<0.05),the proportion of abnormal behavior of each administration group were lower than that in the model group(all P<0.05),the expressions of NF-κB mRNA,ICAM-1 mRNA and protein were lower than those in the model group(all P<0.05),the levels of serum IL-6,IL-1βand CRP were lower than those in the model group(all P<0.05);all above changes in positive group were more significant than those in low and middle dose Tenghuang Jiangu tablets groups(all P<0.05).Conclusion:Tenghuang Jiangu Tablet can effectively improve bone metabolism and behavior in KOA model rats at acute exacerbation stage,and its main mechanism may be related to down-regulation of NF-κB,ICAM-1 expressions and inflammatory factors.
