摘要
为鉴定云南玉米穗腐病主要致病菌、研究其共生真菌群落结构以及了解主要致病菌的流行特征,从云南采集病原样本进行分离纯化,对主要致病菌进行形态学鉴定和致病性测定;并对样本中真菌总DNA的rDNA-ITS序列进行SMRT测序和生物信息学分析。最后依据从文献中收集的中国21个省份穗腐病致病菌的分离鉴定结果,利用空间插值分析推测主要致病菌在中国的分布。研究结果表明,云南玉米穗腐病主要致病菌为拟轮枝镰孢菌(Fusarium verticillioides),其共生真菌群落中弯曲单胞瓶霉(Phialemonium curvatum)、季也蒙毕赤酵母(Meyerozyma caribbica)等稳定存在但在不同患病部位相对丰度不同;镰孢菌属不同的种在中国呈现一定的地域性分布。拟轮枝镰孢菌可能与少数共生真菌存在稳定的互利关系,其在云南的分布可能与玉米品种易感和现代物流导致的扩散有关。
In order to identify the main pathogens of maize ear rot in Yunnan,and the structure of the pathogenic fungi symbiotic community and its epidemic characteristics,pathogens were isolated and purified from the maize ear rot samples collected from Yunnan and were identified based on their morphological and pathogenicity tests.The rDNA-ITS of the fungal pathogen were sequenced(SMRT sequencing)and bioinformatics was analyzed.Based on the pathogens’occurring in 21 provinces in China,the spatial interpolation analysis was used to infer the distribution of maize ear rot’s main pathogens in China.The results showed that Fusarium verticillioides was the main pathogen of maize ear rot in Yunnan.In its symbiotic fungal community,Phialemonium curvatum and Meyerozyma caribbica occurred in most samples,but their relative abundance varied with locations,compared with the consistent distribution of the different species of Fusarium.The main pathogen F.verticillioides might have a mutual benefit relationship with a few other symbiotic fungi,and the distribution of F.verticillioides in Yunnan could be related to the susceptibility of maize varieties and the spreading caused by modern logistics.
