摘要
目的:建立一种简单的高效液相色谱法用于测定人血清中替加环素的浓度。方法:以盐酸米诺环素为内标,乙腈沉淀蛋白法处理样品。色谱柱为KromasilC18柱(4.6mm×150mm,5μm);以乙腈-磷酸盐缓冲液(24∶76,v/v,pH=3.0)为流动相,流速1mL·min^(-1);检测波长246nm;进样量50μL。结果:人血清中替加环素的校准曲线在0.10~5.00μg∙mL^(-1)浓度范围内线性关系良好。低、中、高质量浓度质控样品的日内和日间精密度RSD均<10%,提取回收率在99.43%~116.68%。结论:该方法简便、可靠、灵敏度高,已成功应用于我院接受替加环素治疗患者的样本分析。
Objective:To establish a simple high performance liquid chromatography(HPLC)method for the determination of tigecycline(TGC)in human serum.Methods:The samples were treated by acetonitrile precipitation protein method with minocycline hydrochloride as internal standard.The chromatographic column was Kromasil C18 column(4.6 mm×150 mm,5μm).The mobile phase consisted of acetonitrile phosphate buffer(24∶76,v/v,pH=3.0).The flow rate was 1 mL∙min^(-1) and detection wavelength was 246 nm.Injection volume was 50μL.Results:The calibration curve of TGC in human serum has a good linearity in the concentration range of 0.10–5.00μg∙mL^(-1).The intraday and interday precision RSD of low,medium,high-dose quality control samples were<10%respectively.Extraction recovery was 99.43%–116.68%.Conclusion:This method was simple and reliable with high sensitivity,which was successfully applied for the determination of TGC in patients of our hospital.
