摘要
目的探讨miR-612过表达对宫颈癌SiHa细胞增殖、迁移和侵袭的影响。方法选择宫颈癌SiHa细胞,利用Lipofectamine2000分别将miR-612模拟物、miR-612 NC转染至SiHa细胞,并分为miR-612 mimic组、miR-612 mimic-NC组和空白对照组。采用qRT-PCR法检测各组SiHa细胞miR-612的表达水平;CCK-8法以及流式细胞术分别检测各组细胞的增殖情况和细胞周期;细胞划痕实验以及Transwell实验分别检测各组细胞的迁移和侵袭能力。结果与miR-612 mimic-NC组和空白对照组比较,miR-612 mimic组miR-612的表达水平增高(P均<0.05),提示转染成功。转染48 h后,miR-612 mimic组在各时段的增殖活性较其他两组明显降低(P均<0.05);miR-612 mimic组G0/G1期细胞的比例较其他两组增加(P均<0.05),S期细胞的比例较其他两组减少(P<0.05);miR-612 mimic组的相对划痕宽度较其他两组增加(P均<0.05),细胞侵袭数目较其他两组减少(P均<0.05)。结论过表达miR-612能使宫颈癌SiHa细胞的增殖活性降低,使G0/G1期的细胞比例增加,使SiHa细胞的迁移和侵袭能力均受到抑制。
Objective To investigate the effect of miR-612 overexpression on proliferation,migration and invasion of cervical cancer SiHa cells.Methods Cervical cancer SiHa cells were selected and transfected with miR-612 mimics and miR-612 NC into SiHa cells by Lipofectamine2000,respectively.The cells were divided into the miR-612 mimic group,the miR-612-NC mimic group and the blank control group.The expression level of miR-612 in SiHa cells in each group was determined by qRT-PCR.Cell proliferation and cell cycle were detected by CCK-8 and flow cytometry.Cell scratch test and Transwell test were used to detect the migration and invasion ability of cells in each group.Results Compared with the miR-612 mimic-NC group and the blank control group,the expression level of miR-612 mimicry group was significantly increased(P all<0.05),indicating successful transfection.After transfection for 48 h,the proliferation activity of the miR-612 mimic group at each time period was significantly reduced compared with that of the other two groups(P all<0.05).The proportion of G0/G1 phase cells in the miR-612 mimic group increased compared with the other two groups(P all<0.05),and the proportion of cells in the S phase decreased compared with the other two groups(P all<0.05).The relative scratch width ratio of the miR-612 mimic group was significantly increased compared with the other two groups(P all<0.05),and the number of cell invasions was significantly reduced compared with the other two groups(P all<0.05).Conclusion Overexpression of miR-612 can reduce the proliferation activity of cervical cancer SiHa cells,increase the proportion of cells in G0/G1 phase,and inhibit the migration and invasion of SiHa cells.
