摘要
目的:探讨中药香薷75%乙醇提取物对小鼠内脏痛行为的影响及在体外对瞬时受体电位香草酸亚型1(TRPV1)通道的影响。方法:在福尔马林诱导的急性内脏痛模型小鼠上进行行为学实验,观察香薷醇提物干预后小鼠疼痛行为变化;运用全细胞膜片钳技术检测不同浓度的香薷醇提取物对瞬染人源TRPV1的HEK293细胞(TRPV1/HEK293细胞)诱导产生的跨膜电流,证明香薷醇提物对TRPV1的激活作用。结果:与模型组比较,各给药组小鼠疼痛行为频次均显著降低(P<0.01);与细胞外液比较,3 g/L香薷醇提物能诱导TRPV1/HEK293细胞产生显著跨膜电流(P<0.01),该跨膜电流与TRPV1激动剂辣椒素诱发的电流在电流密度和电流-电压关系方面均相似;香薷醇提物在3、10 g/L浓度时均可使TRPV1/HEK293细胞产生显著跨膜电流(P<0.01),且10 g/L香薷醇提物诱导产生的跨膜电流显著大于3 g/L(P<0.01)。结论:中药香薷醇提取物中含有TRPV1受体激动剂,香薷的止痛机制可能与其激活热敏通道TRPV1有关。
Objective:To explore the effects of 75%ethanol extract of Herba Mosla on visceral pain behavior in mice and TRPV1 channel in vitro.Methods:Behavioral experiments were carried out in formalin-induced acute visceral pain model mice to observe the changes of pain behavior after the intervention of ethanol extract of Herba Mosla.The whole-cell patch clamp technique was used to detect the transmembrane current induced by ethanol extract of Herba Mosla at different concentrations on HEK293 cells(TRPV1/HEK293 cells)transiently infected with human TRPV1,which proved the activation effect of Herba Mosla ethanol extract on TRPV1.Results:The frequency of pain behavior in mice was significantly decreased after pretreatment with each administration groups compared with that in the model group(P<0.01).Compared with the extracellular solution,3 g/L ethanol extract of Herba Mosla could induce obvious transmembrane current in TRPV1/HEK293 cells(P<0.01),which was similar to the current induced by capsaicin,a TRPV1 agonist,in terms of current density and currentvoltage relationship;the ethanol extract of Herba Mosla at 3 and 10 g/L could induce obvious transmembrane current in TRPV1/HEK293 cells(P<0.01),and the transmembrane current induced by 10 g/L ethanol extract of Herba Mosla was greater than 3 g/L(P<0.01).Conclusion:The ethanol extract of Herba Mosla contains TRPV1 receptor agonists,and the analgesic mechanism of Herba Mosla may be related to the activation of TRPV1.
