麻杏石甘汤对流感病毒肺部感染模型肺组织TLR2/4-MyD88信号通路蛋白表达的影响 被引量:4

Effects of Maxing Shigan Decoction on TLR2/4-MyD88 signaling pathway protein expression in lung tissue of influenza virus pulmonary infection model

摘要 目的:从肺组织TLR2/4-MyD88信号通路蛋白表达水平探索麻杏石甘汤抗流感病毒效应机制。方法:经鼻腔接种建立流感病毒肺部感染模型,设正常对照组、模型对照组、奥司他韦组、抗病毒口服液组、麻杏石甘汤组。各治疗组经灌胃给药3、7 d,常规法检测肺指数、肺指数抑制率;HE染色法观察肺组织的病理变化;免疫组织化学法和免疫印迹法检测肺组织TLR2、TLR4、MyD88、NF-κB蛋白表达水平。结果:与正常对照组比较,模型对照组肺指数显著升高(P<0.05);与模型对照组比较,给药治疗3 d的麻杏石甘汤组肺指数显著降低(P<0.05)。与正常对照组比较,模型对照组小鼠肺组织充血水肿,大量炎症细胞浸润;与模型对照组比较,给药治疗3、7 d的麻杏石甘汤组小鼠肺组织病理损伤得到明显改善。免疫组化结果显示:给药3 d,与正常对照组比较,模型对照组肺组织TLR2、TLR4、NF-κB蛋白表达水平显著升高(P<0.05);与模型对照组比较,麻杏石甘汤组肺组织内TLR2、TLR4、NF-κB蛋白表达水平显著下降(P<0.05);给药7 d,与正常对照组比较,模型对照组肺组织内MyD88蛋白表达水平显著升高(P<0.05);与模型对照组比较,麻杏石甘汤组肺组织内MyD88蛋白表达水平显著下降(P<0.05)。免疫印迹法显示:给药3 d,与正常对照组比较,模型对照组肺组织内TLR2、TLR4、MyD88、NF-κB蛋白表达水平显著升高(P<0.05);与模型对照组比较,麻杏石甘汤组肺组织各蛋白表达水平显著降低(P<0.05);给药7 d,各组间差异无统计学意义。结论:麻杏石甘汤可能是通过调节TLR2/4-MyD88信号通路相关蛋白的表达水平而发挥抗流感病毒作用。 Objective:To explore the mechanism of Maxing Shigan Decoction(MXSGD)on influenza virus from the expression level of TLR2/4-MyD88 signaling pathway protein in lung tissue.Methods:The pulmonary infection model of influenza virus was established by intranasal inoculation.The experiment was divided into normal control group,model control group,oseltamivir group,antiviral oral liquid group,MXSGD group.The lung index and lung index inhibition rate were detected by routine method;the pathological changes of lung tissue were observed by HE staining;the protein expression levels of TLR2,TLR4,MyD88 and NF-κB were detected by immunohistochemistry and Western Blot.Results:Compared with the normal control group,the lung index of model control group was significantly increased(P<0.05).Compared with the model control group,the lung index of MXSGD group decreased significantly(P<0.05).Compared with the normal control group,the lung tissue of the model control group was congested and edematous,and a large number of inflammatory cells were infiltrated.Compared with the model control group,the pathological damage of lung tissue in MXSGD group was significantly improved after 3 days and 7 days of treatment.Immunohistochemical results showed that after 3 days of administration,the expression levels of TLR2,TLR4 and NF-κB protein in lung tissues of mice in model control group were significantly increased compared with normal group(P<0.05),while the expression levels of TLR2,TLR4 and NF-κB protein in MXSGD group were significantly decreased compared with that in model control group(P<0.05).After 7 days of administration,the expression level of MyD88 protein in the lung tissues of mice in the model control group was significantly increased compared with the normal group(P<0.05),and the expression level of MyD88 protein in the lung tissues of mice in the MXSGD group was significantly decreased compared with the model control group(P<0.05).Western Blot showed that:3 days after administration,compared with normal group,the expression le
