摘要
目的探讨复方丹参滴丸(compound danshen dripping pills,CDDP)对肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)诱导的人牙周膜成纤维细胞(human periodontal ligament cells,hPLFCs)凋亡和凋亡相关蛋白的影响。方法体外培养hPLFCs,MTT法检测TNF-α对细胞增殖的影响,检测CDDP对于TNF-α诱导hPLFCs增殖的影响。Annexin V/PI检测CDDP对于TNF-α诱导hPLFCs凋亡率的变化。Western blot检测CDDP对于TNF-α诱导的hPLFCs凋亡相关蛋白p53、Bax及Bcl-2的影响。结果结果显示,与对照组比较,hPLFCs细胞经TNF-α处理后,细胞增殖显著下降,细胞凋亡率显著增加,p53和Bax表达水平明显升高,Bcl-2表达水平明显降低。与TNF-α组比较,hPLFCs细胞经剂量依赖性CDDP处理后,细胞增殖逐渐升高,细胞凋亡率逐渐下降,p53和Bax表达逐渐下降,Bcl-2表达逐渐升高。结论CDDP可以剂量依赖性降低TNF-α诱导的hPLFCs凋亡率及凋亡相关蛋白的表达水平,对抑制hPLFCs凋亡有着重要意义。
Objective To investigate the effects of compound danshen dripping pills(CDDP)on tumor necrosis factor-α(TNF-α)induced apoptosis and apoptosis-related proteins in human periodontal ligament cells(hPLFCs).Methods hPLFCs were cultured in vitro,and the effect of TNF-αon cell proliferation was detected by MTT method.The effect of CDDP on hPLFCs proliferation which induced by TNF-αwas detected by MTT method.Annexin V/PI was used to detect the effect of CDDP on hPLFCs apoptosis rate which induced by TNF-α.Western blot was used to detect the effects of CDDP on TNF-α-induced hPLFCs apoptosis related proteins p53,Bax and Bcl-2.Results The results showed that compared with the control group,the proliferation of hPLFCs was significantly decreased,the apoptosis rate was significantly increased,the expression of p53 and Bax was gradually increased,and the expression of Bcl-2 was gradually decreased in the TNF-αgroup.Compared with the TNF-αgroup,hPLFCs cells treated with dose-dependent CDDP showed increased proliferation,decreased apoptosis rate,decreased the expression of p53 and Bax,and increased the expression of Bcl-2.Conclusion CDDP could reduce the apoptosis rate and the expression level of apoptosis-related proteins of hPLFCs which induced by TNF-αin a dose-dependent manner,which is of great significance to inhibit the apoptosis of hPLFCs.
