摘要
目的探讨香青兰总黄酮对胃癌MGC-803细胞恶性生物学行为的影响及巨核细胞白血病因子(MKL)1/半胱氨酸天冬氨酸蛋白水解酶(Caspase)-3活性和凋亡抑制因子(CAAP)1通路的作用。方法体外培养胃癌MGC-803细胞,对照组不进行处理,香青兰总黄酮低剂量组、香青兰总黄酮中剂量组、香青兰总黄酮高剂量组分别给予浓度为25、50、100 mg/L的香青兰总黄酮,曲妥珠单抗组给予浓度为10 mg/L的曲妥珠单抗,继续培养24 h后检测细胞增殖率、细胞凋亡率、细胞侵袭和迁移能力,同时检测胃癌MGC-803细胞中MKL1、CAAP1、基质金属蛋白酶(MMP)-2、Caspase-3 mRNA和蛋白水平。结果与对照组比较,各香青兰总黄酮剂量组细胞增殖率、细胞侵袭数、迁移距离、MKL1、CAAP1、MMP-2、Caspase-3 mRNA和蛋白水平显著降低,细胞凋亡率显著增加,且呈剂量依赖性,但效果不如曲妥珠单抗组(P<0.05)。结论香青兰总黄酮可以降低胃癌MGC-803细胞增殖、抑制其侵袭和迁移、同时促进细胞凋亡,其机制可能与MKL1/CAAP1通路的抑制有关。
Objective To investigate the effect of total flavonoids of dracocephalum moldavica on malignant biological behavior of gastric cancer MGC-803 cells and the role of megakaryocytic leukemia(MKL)1/Caspase activity and inhibitor of apoptosis(CAAP)1 pathway.Methods Gastric cancer MGC-803 cells were cultured in vitro,and the control group was left untreated.The total flavonoids of dracocephalum moldavica low dose group,the total flavonoids of dracocephalum moldavica middledose group and the total flavonoids of dracocephalum moldavica high dose group were given final concentrations of 25,50,100 mg/L total flavonoids of dracocephalum moldavica,trastuzumab group was given 10 mg/L trastuzumab.After 24 hours of culture,the cell proliferation rate,apoptosis rate,cell invasion and migration ability were tested;the mRNA and protein levels of MKL1,CAAP1,MMP-2,Caspase-3 in gastric cancer MGC-803 cells were detected simultaneously.Results Compared with the control group,the proliferation rate,cell invasion number,migration distance,MKL1,CAAP1,MMP-2 and Caspase-3 mRNA and protein levels were decreased in gastric cancer MGC-803 cells of each total flavonoids of dracocephalum moldavica groups,and the cell apoptosis rate was increased,and the effect was dose-dependent in total flavonoids of dracocephalum moldavica groups,but the effect of each index was not as good as that of trastuzumab group(P<0.05).Conclusions Total flavonoids of dracocephalum moldavica could reduce the proliferation,invasion and migration of gastric cancer MGC-803 cells,and promote cell apoptosis.The mechanism may be related to the inhibition of MKL1/CAAP1 pathway by total flavonoids of dracocephalum moldavica.
