基于微流控芯片的毕赤酵母微胶囊收集系统构建

摘要 微流控技术已成为目前发酵工程中筛选和表征微生物性能的重要方法。本研究从微流控技术的理论出发,将计算流体力学、颗粒-流体两相流动和牛顿运动定律理论用于粒子追踪模型,建立了毕赤酵母微胶囊收集过程的数学模型。借助仿真软件模拟了毕赤酵母微胶囊的收集过程,设计了微流控芯片中收集器结构,并进行了进一步的数值模型模拟,最后搭建了用于分析毕赤酵母微胶囊的实验平台。本研究为研究毕赤酵母单细胞表达重组蛋白奠定了一定基础。 Microfluidics has become an important method for screening and characterizing the performance of microorganisms in fermentation engineering.In this project,a mathematical model of the microcapsule collection process was developed according to the theory of microfluidics,particle-fluid two-phase flow and Newton’s laws of motion solid mechanics,using particle tracking models.The collection process of Pichia pastoris microcapsules was simulated with the help of simulation software,the collector structure in the microfluidic chip was designed,and further numerical model simulations were carried out.Then,the collector was designed and set up for experimental verification of microcapsule collection and analysis.This research lays a foundation for research about recombinant protein expression by recombinant K.phaffii.
