摘要
目的检测miRNA-126、miRNA-26a在食管癌患者中的表达水平,并分析miRNA-126、miRNA-26a表达情况与食管癌临床特征的关系。方法收集113例食管癌手术切除患者的食管癌组织和癌旁组织,分析miRNA-126和miRNA-26a在食管癌组织和癌旁组织中的相对表达量及相关性,采用受试者工作特征(ROC)曲线分析miRNA-126、miRNA-26a的预测价值。结果食管癌组织中miRNA-126和miRNA-26a相对表达量均明显低于癌旁组织,差异均有统计学意义(P﹤0.01)。ROC曲线分析结果显示,当约登指数最大时,对应的miRNA-126和miRNA-26a为0.92和2.03,可作为诊断食管癌的临界值。miRNA-126和miRNA-26a在食管癌组织中的表达水平呈正相关(r=0.757,P﹤0.01)。不同分化程度、TNM分期及淋巴结转移情况的食管癌患者中miRNA-126和miRNA-26a表达情况比较,差异均有统计学意义(P﹤0.01)。结论 miRNA-126、miRNA-26a在食管癌患者中呈低表达,且与食管癌分化程度、TNM分期及淋巴结转移有关,可反映病情严重程度和预后。
Objective To detect the expression levels of miRNA-126 and miRNA-26a in patients with esophageal cancer,and to analyze the relationship between the expression of two markers and patients’clinical characteristics.Method The esophageal cancer tissues and adjacent tissues of 113 patients with esophageal cancer surgical resection were collected,and the relative expression and correlation of miRNA-126 and miRNA-26a in esophageal cancer tissues and paracancerous tissues were also analyzed.The receiver operating characteristic(ROC)curve was used to analyze the predictive value of miRNA-126 and miRNA-26a.Result The relative expression levels of miRNA-126 and miRNA-26a in esophageal cancer tissues were significantly lower than those in the adjacent tissues,and the differences were statistical-ly significant(P<0.01).The results of ROC curve analysis showed that when the largest Youden index was selected,the corresponding values of miRNA-126 and miRNA-26a were 0.92 and 2.03,which could be used as the cut value for esoph-ageal cancer diagnoses.The expression levels of miRNA-126 and miRNA-26a in esophageal cancer tissues were positive-ly correlated(r=0.757,P<0.01).There were significant differences in the expression of miRNA-126 and miRNA-26a in esophageal cancer patients with different differentiation grades,TNM stages,and lymph node metastasis(P<0.01).Conclusion The indicators miRNA-126 and miRNA-26a were down-regulation in patients with esophageal cancer and were related to the differentiation grade,TNM stage,and lymph node metastasis,which could be used to reflect the severi-ty and prognosis of the disease.
