基于GEO芯片评价乳品中胰岛素生长因子-Ⅰ、维生素D3和雌二醇联合对乳腺癌标志因子效应的研究 被引量:1

摘要 多项研究表明雌二醇(E2)、维生素D_(3)(VD_(3))和胰岛素生长因子-Ⅰ(IGF-Ⅰ)均对乳腺病发生及身体发育机能调控等具有潜在影响,但目前缺乏三者联合效应的研究报道。本研究主要基于生物信息学方法,分析乳品中E2、VD_(3)、IGF-Ⅰ联合效应及其对乳腺癌标志因子的调控机制,为科学摄取乳品提供参考依据。依据美国国立生物技术信息中心(NCBI)人乳腺癌细胞(MCF-7)的3套GEO芯片(GSE160497、GSE27220、GSE145787),使用cytoscape3.7.2筛选关键基因,应用R软件进行GO/KEGG通路富集分析,挖掘E2、VD_(3)、IGF-Ⅰ联合效应,探究三者对乳腺癌标志因子的调控机制。结果表明E2、VD_(3)、IGF-Ⅰ共同作用DNA复制、染色体合成等促进机体生长发育信号通路,可使乳腺癌标志因子MUC1表达沉默。E2单独作用可使微小染色体维持蛋白7、核糖体RNA加工蛋白等基因显著下调,可促进MUC1过表达。认为E2可与VD_(3)、IGF-Ⅰ互作维持平衡机体代谢,但过量E2可能激活MUC1乳腺癌标志因子通路,因此特殊人群正常摄入乳品可满足营养及生理需要,但额外补充雌激素应慎重。 Breast cancer has become the most prevalent cancer worldwide,and several studies have shown that estradiol(E2),vitamin D_(3)(VD_(3))and insulin growth factor 1(IGF-Ⅰ)all have potential effects on the development of breast disease and regulation of body developmental functions,but there are still no reports on the combined effects of the three.In this study,we analyzed the combined effects of E2,VD_(3) and IGF-Ⅰ in dairy products and their regulatory mechanisms on breast cancer markers based on a bioinformatics approach to provide a reference for scientific intake of dairy products.According to 3 sets of GEO chips(GSE160497,GSE27220,GSE145787)of human breast cancer cells(MCF-7)of the National Center for Biotechnology Information(NCBI),use cytoscape 3.7.2 to screen key genes,and use R software for GO/KEGG pathway enrichment Analyze,explore the combined effects of E2,VD_(3),and IGF-Ⅰ,and explore the regulatory mechanisms of the three on breast cancer marker factors.E2,VD_(3),and IGF-Ⅰ work together to promote the growth and development of the body,such as DNA replication and chromosome synthesis,which can silence the expression of breast cancer marker MUC1.E2 alone can significantly down-regulate genes such as microchromosome maintenance protein 7,ribosomal RNA processing protein,and promote the overexpression of MUC1.E2 can interact with VD_(3) and IGF-Ⅰ to maintain a balanced body metabolism.However,excessive E2 may activate the pathway of MUC1 breast cancer marker factors.Therefore,the daily diet of special populations should be cautiously ingesting foods rich in estrogen.
