摘要
目的探索基于熵权TOPSIS法综合评价血管性痴呆(vascular dementia,VD)大鼠学习记忆能力。方法24只雄性SD大鼠随机分为实验组(VD,分次结扎双侧颈总动脉)和对照组(假手术)。造模8周后采用Morris水迷宫检测大鼠学习记忆能力,熵权法对水迷宫指标赋予权重,TOPSIS模型计算大鼠学习记忆能力综合评价得分并排序。结果熵权法赋予可纳入评价模型的潜伏期等23项指标的权重为0.0016~0.1824,指标权重排名前三位的分别是:站台穿越次数(0.1824)、总时间(0.1229)、潜伏期(0.1067)。指标权重排名后三位的分别是:中央活动时间(0.0043)、第三象限时间(0.0037)、搜索策略(0.0016)。TOPSIS法可得到两组大鼠Morris水迷宫学习记忆能力综合评价得分为0.0243~0.0641,排名前三位分别为:D012(0.0641)、D007(0.0524)、D010(0.0510),排名后三位分别为:S004(0.0309)、S010(0.0290)、S009(0.0243),对照组综合得分相较于实验组升高(P<0.05)。结论熵权TOPSIS法能更全面客观地反映VD大鼠学习记忆能力水平,可为认知功能障碍大鼠模型是否构建成功以及药物等因素对大鼠学习记忆能力的影响等研究提供参考。
Objective To decipher the application of entropy-weight based TOPSIS method in the comprehensive evaluation of the learning and memory ability of rats with vascular dementia(VD).Methods Twentyfour male SD rats were randomly divided into the experimental group and the control group.Rats with VD in the experimental group were induced by bilateral common carotid artery occlusion in stages,while control rats were treated with pseudo-surgery.8weeks after the surgery,the learning and memory of rats were assessed through Morris water maze.The entropy method was used to assign weights to the water maze indicators and TOPSIS model was used to score and evaluate the learning and memory ability of rats.Results The weights of 23indicators which could be included in the assessment model and assigned by entropy method ranged from 0.0016to 0.1824.The top three weighted indicators were numbers that rats crossed over the platform(0.1824),total time spending(0.1229)and the latency(0.1067),while the bottom three indicators were central activity time(0.0043),third quadrant time(0.0037)and search strategy(0.0016).The scores evaluated by TOPSIS method on learning and memory abilities of the rats in both groups through Morris water maze ranged from 0.0243to 0.0641,with the top three of D012(0.0641),D007(0.0524),D010(0.0510),and the bottom three of S004(0.0309),S010(0.0290),S009(0.0243).The overall score of rates in the control group was significantly higher than that of those in the experimental group(P<0.05).Conclusions Entropy-weight based TOPSIS method can comprehensively and objectively evaluate the learning and memory of VD rats,which can provide reference for the successful construction of the cognitive dysfunction rat model and assessment of the influence of drugs and other factors on the learning and memory ability of rats.
