摘要
基于网络药理学探讨药食同源玫瑰花的有效化合物及其潜在药理及保健作用机制。利用中药系统药理学技术平台(TCMSP)和蛋白质相应作用平台(STRING),过滤玫瑰花的有效化合物及对应作用靶点;运用Cytoscape 3.7.0软件建立并分析玫瑰花有效化合物-作用靶点网络;利用STRING数据库平台和Cytoscape 3.7.0软件建立并分析玫瑰花有效化合物作用靶点PPI网络;利用Bioconductor对作用靶点进行生物功能注释(GO)和KEGG通路富集分析。结果显示:玫瑰花中的番茄红素、Rubixanthin、Rugosin A、β-谷甾醇、豆甾醇、Cyanine、维生素E、去甲氧基茵陈色原酮、β-胡萝卜素和槲皮素10个有效化合物可能通过AGE-RAGE、TNF、IL-17和HIF-1等161条信号通路作用于PTGS2、NCOA2、PTGS1、BCL2、CASP9、JUN、CASP3和CASP8等154个作用靶点,发挥抗氧化、抗肿瘤、抗炎、抗病毒、免疫调节和保肝等药理作用以及降糖、降脂、抗衰老及改善皮肤等保健功能。玫瑰花发挥药理及保健作用具有多成分、多靶点和多途径的特点,研究结果可为玫瑰花药用与食用价值的进一步研究与开发提供理论基础与科学依据。
To investigate the potential mechanism of effective components, pharmacological action and health care function of Rosae rugosae flos based on network pharmacology.TCMSP and STRING database were used to filter the effective components and related targets of R.rugosae flos.Cytoscape 3.7.0 software was used to establish and analyze the effective component-target network R.rugosae flos.The PPI network was constructed and analyzed by STRING database and Cytoscape 3.7.0 software, and the Bioconductor was used to analyze the GO function and KEGG pathway enrichment of the targets.The results showed that lycopene, rubixanthin, rugosin A,β-sitosterol, sitosterol, cyanine, vitamin-e, demethoxycapillarisin, β-carotene and quercetin of R.rugosae flos acted on 154 targets including PTGS2,NCOA2,PTGS1,BCL2,CASP9,JUN,CASP3 and CASP8 through 161 pathways including AGE-RAGE,TNF,IL-17 and HIF-1,exerting the pharmacological functions involving anti-oxidation, anti-tumor, anti-inflammatory, anti-virus, immune regulation, liver protection, and health care functions involving hypoglycemic, lipid-lowering, anti-aging, skin improvement.R.rugosae flos has the characteristics of multi-component, multi-target and multi-channel in its health care function and pharmacological action.The results can provide theoretical basis and scientific basis for further research and development of R.rugosae flos.
