摘要
目的:初步探讨长链非编码RNA LINC00511和血管内皮生长因子(VEGF)调控肺腺癌增殖、侵袭和迁移的机制。方法:qRT-PCR检测肺腺癌组织样本中LINC00511和VEGF的表达;qRT-PCR和Western Blot检测肺腺癌细胞中LINC00511和VEGF的表达;设计并合成shRNA敲低LINC00511和VEGF后,通过CCK8和Transwell实验,检测肺腺癌细胞的增殖、侵袭和迁移;通过qRT-PCR和Western Blot,检测敲低LINC00511后VEGF的表达;敲低LINC00511的同时过表达VEGF,通过CCK8和Transwell实验,检测肺腺癌细胞的增殖、侵袭和迁移。结果:LINC00511和VEGF在肺腺癌的组织样本和肺腺癌细胞A549中,均显著高表达;敲低LINC00511和VEGF都能显著抑制肺腺癌细胞的增殖、侵袭和迁移;敲低LINC00511,能显著抑制VEGF的表达水平;敲低LINC00511的同时过表达VEGF,能恢复肺腺癌细胞的增殖、侵袭和迁移能力。结论:LINC00511通过血管内皮生长因子VEGF调控肺腺癌的增殖、侵袭和迁移。
Objective: To preliminarily investigate the mechanism of long non-coding RNA LINC00511 and vascular endothelial growth factor(VEGF)in regulating proliferation, invasion and migration of lung adenocarcinoma. Methods: qRT-PCR was used to detect the expression of LINC00511 and VEGF in lung adenocarcinoma tissues;qRT-PCR and Western Blot were performed to measure the expression of LINC00511 and VEGF in lung adenocarcinoma cells;CCK8 and transwell assay were used to detect the proliferation, invasion and migration of lung adenocarcinoma cells. After shRNA were designed and synthesized to knock down the levels of LINC00511 and VEGF;qRT-PCR and Western Blot were conducted to quantified the expression of VEGF after down-regulation of LINC00511;CCK8 and transwell assays were used to examine the proliferation, invasion, and migration of lung adenocarcinoma cells after down-regulation of LINC00511 and over-expression of VEGF. Results:LINC00511 and VEGF were significantly highly expressed in tissue samples of lung adenocarcinoma and lung adenocarcinoma cell A549;knockdown of LINC00511 and VEGF could significantly inhibit the proliferation, invasion and migration of lung adenocarcinoma cells;knockdown of LINC00511 could significantly inhibit the expression level of VEGF;knockdown of LINC00511 along with overexpression of VEGF could restore the proliferation, invasion and migration abilities of lung adenocarcinoma cells. Conclusion: LINC00511 could regulate the proliferation, invasion, and migration of lung adenocarcinomas through VEGF.
