摘要
以结球红菊苣地上部分为原料,采用复合酶法水解浸提菊苣多糖,再经Sevag法脱蛋白、大孔树脂脱色、DEAE纤维素柱层析等过程获得高纯度菊苣多糖。通过高效液相色谱、红外光谱及核磁等技术对多糖组分进行分析表征;最后通过测定多糖组分清除DPPH、ABTS和羟自由基的能力,评价其体外抗氧化活性。结果表明:经分离纯化,得到多糖组分CCP-1、CCP-2。CCP-1由果糖、葡萄糖组成,其单糖摩尔百分比(%)约为82.13:17.87,CCP-2含有果糖、葡萄糖、半乳糖,其单糖摩尔百分比(%)约为93.19:2.57:4.24。CCP-1的分子质量为3482 u,CCP-2的分子质量为2879 u。经红外光谱分析显示,CCP-1以α-糖苷键和β-糖苷键相连的吡喃糖环多糖,CCP-2以α-糖苷键相连的吡喃糖环多糖。核磁分析结果与红外光谱分析一致。CCP-1、CCP-2表现出一定的体外抗氧化活性,当浓度为4 mg/mL时,CCP-1对DPPH自由基、ABTS自由基和羟自由基的清除率分别为43.21%、36.57%和40.52%,CCP-2的清除能力分别为57.90%、52.29%和53.67%。该研究结论为结球红菊苣多糖进一步深入研究、功能性食品或药品的开发提供理论依据。
In this paper,Cichoricum intybus var.foliosum Hegi.was used as raw material to hydrolyze and extract Cichoricum intybus var.foliosum Hegi.polysaccharides by complex enzymatic hydrolysis,and then deproteinized by sevag method,macroporous resin decolorization,DEAE cellulose column chromatography and other processes to obtain high-purity chicory polysaccharides.The polysaccharide components were analyzed and characterized by high performance liquid chromatography,infrared spectroscopy,and nuclear magnetic technology.Finally,the ability of the polysaccharide components to scavenge DPPH,ABTS and hydroxyl free radicals was measured to evaluate its in vitro antioxidant activity.The results showed that:after separation and purification,polysaccharide components CCP-1 and CCP-2 were obtained.CCP-1 is composed of fructose and glucose,and its monosaccharide molar percentage (%) is about 82.13:17.87.CCP-2 contains fructose,glucose,and galactose,and its monosaccharide molar percentage (%) is about 93.19:2.57:4.24.The molecular weight of CCP-1 is 3482 u,and the molecular weight of CCP-2 is 2879 u.Infrared spectroscopy analysis shows that CCP-1 is a pyranose ring polysaccharide connected by α-glycosidic bonds and β-glycosidic bonds,and CCP-2 is a pyranose ring polysaccharide connected by α-glycosidic bonds.The NMR analysis results are consistent with the infrared spectroscopy analysis.CCP-1 and CCP-2 show certain in vitro antioxidant activity.When the concentration is 4 mg/mL,the scavenging rates of CCP-1 on DPPH free radicals,ABTS free radicals and hydroxyl free radicals are 43.21%,36.57% and 40.52%,respectively.The clearance capacity of CCP-2 is 57.90%,52.29% and 53.67%,respectively.It provides a theoretical basis for further in-depth research on the polysaccharides of Cichoricum intybus and the development of functional foods or medicines.
