摘要
目的:应用网络药理学分析对“香附-郁金”药对治疗乳腺增生病的作用机制进行探讨与分析。方法:采用中药系统药理学数据库与分析平台(TCMSP)对香附、郁金的有效活性成分进行筛选,并提取其作用靶点,再利用GeneCards数据库获取乳腺增生病相关基因。映射疾病靶点与药物靶点,通过STRING在线数据库构建关键蛋白之间所产生的相互作用关系图;Cytoscape 3.8.0建立“药物-有效成分-作用靶点”网络图,并对关键靶点进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)富集分析。结果:共获得“香附-郁金”药对33个活性成分,包括异鼠李素、槲皮素、豆甾醇等,与疾病潜在靶点取交集后得到88个共同作用靶点,包括ESR1、EGFR、PGR等,GO分析得到104个分子功能、41个细胞组分和1262个生物过程,KEGG通路分析主要涉及119条通路,主要相关通路为PI3K/AKT信号通路。结论:“香附-郁金”药对治疗乳腺增生病多成分、多靶点协同发挥作用机制,可能通过调节人体内激素内分泌水平,从而发挥治疗乳腺增生病的作用,为临床应用奠定了良好的理论基础。
Objective:To explore and analyze the mechanism of“Cyperi Rhizoma-Curcumae Radix”in the treatment of hyperplasia of mammary glands.Methods:The traditional Chinese medicine system pharmacology database and analysis platform(TCMSP)was used to screen the active ingredients of Cyperi Rhizoma-Curcumae Radix,and their targets were extracted,and then the related genes of mammary gland hyperplasia were obtained by using the GeneCards database.Mapping disease targets and drug targets,and the STRING online database was used to construct the interaction diagram between key proteins;Cytoscape 3.8.0 established a“drug-active ingredient-target”network diagram,and analyzed the key targets,gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis.Results:A total of 33 active ingredients of“Cyperi Rhizoma-Curcumae Radix”were obtained,including isorhamnetin,quercetin,stigmasterol,etc.,and 88 common targets were obtained after intersecting with potential disease targets,including ESR1,EGFR,PGR,etc.,104 results were obtained from the GO molecular function analysis,41 results from the cellular component analysis,and 1262 results from the biological process analysis.The KEGG pathway analysis mainly involved 119 pathways,and the main related pathway was the PI3K-AKT signaling pathway.Conclusion:The mechanism of Cyperi Rhizoma-Curcumae Radix in the treatment of hyperplasia of mammary glands is multi-component and multi-target synergy,which may play a role in the treatment of hyperplasia of mammary glands by regulating the hormone endocrine level in human body,and lay a good theoretical foundation for clinical application.
