补肾宁心颗粒多种活性成分同时测定

Simultaneous determination of various active components of Bushenningxin granule

摘要 目的探究补肾宁心颗粒里芍药的主要指标性成分芍药苷、丹参的的主要指标性成分丹酚酸B和苍术的主要指标性成分苍术素的含量测定方法。方法对芍药苷、苍术素、丹酚酸B用HPLC方法测定含量,使用Apollo-C_(18)色谱柱(250mm×4.6mm),利用有机溶剂甲醇提取,选用梯度洗脱的方法,0.2%H_(3)PO_(4)溶液(A)-乙睛(B)为流动相,检测波长分别为230、286、340nm,洗脱速度为1.0ml/min。结果三个成分皆呈现出了良好的线性关系(R^(2)=1.00),分离程度较好,各色谱峰之间无相互干扰。平均回收率为97.45%、97.68%、96.88%(RSD值分别为1.74%、1.50%、0.78%)。结论本文的方法可用于补肾宁心临床前研究及质量控制,能够对芍药苷、苍术素、丹酚酸B3个成分进行含量测定,可为研究提供依据。 Objective To determine the content of paeoniflorin,salvianolic acid B and atractylodes in Bushenningxin granules.Methods The contents of paeoniflorin,atractylodes and salvianolic acid B were determined by HPLC.Apollo-C_(18) column(250mm×4.6mm)was used to extract paeoniflorin with organic solvent methanol.Gradient elution method was used with 0.2%H_(3)PO_(4) solution(A)-ethyl acetate(B)as mobile phase.The detection wavelength was 230nm,286nm,340nm,and elution rate was 1.0ml/min.Results The three components showed a good linear relationship(R^(2)=1.00),and the separation degree was good.There was no mutual interference among the chromatographic peaks.The average recoveries were 97.45%,97.68%and 96.88%(RSD:1.74%,1.50%and 0.78%,respectively).Conclusion This method can be used for the preclinical study and quality control of Bushenningxin,and can be used for the content determination of paeoniflorin,atractylodes and salvianolic acid B,which can provide a basis for the study.
