缓解羊热应激复方中药对小鼠的急性毒性研究 被引量:1

Acute toxicology of compound Chinese herb for alleviating sheep heat stress in mice

摘要 为了验证缓解羊热应激复方中药的安全性,试验根据霍恩氏法,按体重2150,4640,10000,21500 mg/kg的给药剂量给小鼠灌胃,给药后第14天计算小鼠半数致死量(LD;);在最大给药量试验中,试验组最大给药量为按体重120 g/kg给小鼠灌胃给药,对照组小鼠灌胃等体积的生理盐水。试验期间每天观察并记录小鼠的精神状况、呼吸、有无特殊的叫声、毛光洁度、食欲、粪便以及死亡等情况。灌药后第0,1,2,4,7天对各组小鼠进行空腹称重,计算小鼠的增重、采食量、体重增长率;试验结束后,每组随机选取4只小鼠眼球采血后检测血常规和血清生化指标,然后随机选取3只小鼠经颈椎脱臼处死,剖检并观察,计算心脏、肝脏、脾脏、肺脏和肾脏的脏器系数,同时对脏器组织的显微结构进行病理学观察。结果表明:复方中药的剂量增至21500 mg/kg时,小鼠未见死亡,LD;>21500 mg/kg。最大给药量时,小鼠未出现中毒现象,且试验期间试验组和对照组小鼠的采食量、体重和体重增长率也差异不显著(P>0.05);虽然试验组小鼠的血红蛋白浓度显著高于对照组(P<0.05),但仍在正常参考值范围内,且红细胞数、白细胞数、血小板总数等血常规指标和丙氨酸氨基转移酶活性,尿素氮、葡萄糖浓度等血清生化指标与对照组比较均差异不显著(P>0.05)。此外,剖检未见心脏、肝脏、脾脏、肺脏和肾脏发生病变,试验组和对照组各脏器系数均差异不显著(P>0.05)。脏器组织切片的显微观察显示,试验组小鼠未发现与药物相关的实质性病理变化。说明该复方中药无不良反应,安全可靠。 The purpose of the study was to verify the safety of compound traditional Chinese herb for alleviating sheep heat stress in sheep.According to Horn’s method,mice were given gavage at doses of 2150,4640,10000,and 21500 mg/kg body weight,and the lethal dose(LD;)was calculated on the 14 th day after administration.In the maximum dose test,the experimental group was given 120 g/kg of normal saline by gavage,and the mice in the control group were given equal volume of normal saline by gavage.During the experiment,the mental state,breathing,presence or absence of special calls,hair smoothness,appetite,feces and death of the mice were observed and recorded every day.The mice in each group were weighed on an empty stomach at 0,1,2,4,and 7 days after the injection,and the weight gain,feed intake,and weight growth rate of the mice were calculated.After the experiment,4 mice in each group were randomly selected for blood sampling and blood biochemical indexes were detected.Then three mice were selected to be sacrificed by cervical dislocation,and then subjected to autopsy and observation.The coefficients of organs such as heart,liver,spleen,lung and kidney were counted.Meanwhile,the microstructure of the organs was observed by pathology.The results showed that when the dose of compound Chinese herb increased up to 21500 mg/kg,no dead mice were observed and the LD;>21500 mg/kg.There was no toxicity in mice at the maximum dose,and there were no significant differences in feed intake,body weight and body weight growth rate between experimental group and control group during the experiment(P>0.05).Although the hemoglobin concentration of the experimental group was higher than that of the control group(P<0.05),it was still within the normal range.In addition,conventional blood indexes such as red blood cell number,white blood cell number and platelet number,and serum biochemical indexes such as alanine aminotransferase activity,urea nitrogen and glucose concentration were not significantly different from control groups(P>0.05)
