苏木乙酸乙酯提取物对野百合碱诱导肺动脉高压大鼠肺动脉压、血浆BNP及肺小动脉的影响

摘要 目旳研究苏木乙酸乙酯提取物对野百合碱诱导肺动脉高压大鼠肺动脉压、血浆脑钠肽(BNP)及肺小动脉的影响。方法采用完全随机化的分组方法将50只健康雄性SD大鼠随机分为空白组、模型组、苏木乙酸乙酯高剂量组、苏木乙酸乙酯低剂量组、阿魏酸钠组,每组10只。除空白组外,其余组大鼠采用腹腔注射野百合碱方法制备肺动脉高压模型。实验第15天开始,空白组和模型组给予羧甲基纤维素钠溶液10 mL/(kg·d)灌胃,苏木乙酸乙酯高、低剂量组分别给予苏木乙酸乙酯提取物溶液1472 mg/(kg·d)[生药量2.3 g/(kg·d)]、736 mg/(kg·d)[生药量1.15 g/(kg·d)]灌胃,阿魏酸钠组给予阿魏酸钠0.3 g/(kg·d)灌胃,持续至实验第28天。实验期满检测各组大鼠平均肺动脉压(mPAP),测定血浆BNP水平,HE染色观察肺组织及肺小动脉病理形态学变化,计算管壁厚度/血管直径、管壁面积/血管面积。结果造模各组大鼠mPAP、血浆BNP水平、肺小动脉管壁厚度/血管直径、管壁面积/血管面积均明显高于空白组(P均<0.05),苏木乙酸乙酯高剂量组、苏木乙酸乙酯低剂量组和阿魏酸钠组大鼠上述各指标均明显低于模型组(P均<0.05);苏木乙酸乙酯低剂量组大鼠mPAP、血浆BNP水平均明显低于苏木乙酸乙酯高剂量组(P均<0.05),苏木乙酸乙酯高剂量组和苏木乙酸乙酯低剂量组肺小动脉管壁厚度/血管直径、管壁面积/血管面积均明显低于阿魏酸钠组(P均<0.05)。结论苏木乙酸乙酯提取物能降低肺动脉压力及血浆BNP水平,减轻肺小动脉的病理损伤。 Objective It is to study the effects of hematoxylin ethyl acetate(HEA)extract on pulmonary arterial pressure,plasma brain natriuretic peptide(BNP)and pulmonary arterioles in rats with monocrotaline-induced pulmonary hypertension.Methods Fifty healthy male SD rats were randomly divided into blank group,model group,high-dose HEA group,low-dose HEA group and sodium ferulate group by a completely randomized grouping method,10 rats in each group.Except for the blank group,the rats in the other groups were given intraperitoneal injection of monocrotaline to establish the pulmonary hypertension models.From the 15 th day of the experiment,the blank group and model group were given sodium carboxymethyl cellulose solution 10 mL/(kg·d)by gavage,and the high-dose and low-dose HEA groups were respectively given HEA extract solution 1472 mg/(kg·d)[crude drug amount 2.3 g/(kg·d)],736 mg/(kg·d)[crude drug amount 1.15 g/(kg·d)]by gavage,the sodium ferulate group was given ferulic acid sodium 0.3 g/(kg·d)by gavage,all the treatment were lasted until the 28 th day of the experiment.At the end of the experiment,the mean pulmonary arterial pressure(mPAP)of the rats in each group was detected,the plasma BNP level was determined,the pathological changes of lung tissue and pulmonary arterioles were observed by HE staining,and the wall thickness/vessel diameter and vessel wall area/vessel area were calculated.Results The mPAP,levels of plasma BNP,pulmonary arteriolar wall thickness/vascular diameter,and wall area/vascular area in each modeled group were significantly higher than those in the blank group(all P<0.05).The above indexes of rats in the high-dose and low-dose HEA groups and sodium ferulate group were significantly lower than those in the model group(all P<0.05);the mPAP and levels of plasma BNP in the rats of the low-dose HEA group were significantly lower than those in the the high-dose HEA group(all P<0.05),the pulmonary arteriole wall thickness/vessel diameter,vessel wall area/vessel area in the high-dose and low-dose
