摘要
目的:探讨归芪白术方联合奥沙利铂在IL-6/JAK2/STAT3通路中对MFC胃癌荷瘤小鼠的抑瘤作用及对炎症分子影响。方法:建立MFC胃癌荷瘤小鼠模型,随机分为模型组、奥沙利铂组、奥沙利铂加归芪白术方高、中、低剂量组,10只/组,另选10只健康小鼠作为空白组;各组小鼠经口灌胃给予相应药物,空白组、模型组给予生理盐水,连续治疗14 d。处死小鼠取脾脏、胸腺、肿瘤称重,计算脏器指数及抑瘤率;HE染色观察小鼠瘤体病理形态学变化;ELISA法检测小鼠血清中IL-6含量;RT-qPCR和Western blot分别检测瘤组织中IL-6、JAK2、STAT3 mRNA及IL-6、p-JAK2、p-STAT3蛋白含量;免疫组化法检测肿瘤组织中c-Myc、Cyclin D1蛋白。结果:各治疗组小鼠瘤体重量显著低于模型组(P<0.01),联合用药高、中剂量组瘤体重量明显低于奥沙利铂组(P<0.05,P<0.01);与空白组相比,模型组小鼠的脾指数、胸腺指数降低(P<0.01),与模型组相比,奥沙利铂组小鼠的脾指数、胸腺指数降低(P<0.01),与奥沙利铂组相比,联合用药高剂量组脾指数升高,差异有统计学意义,联合用药高、中剂量组胸腺指数增加,差异有统计学意义(P<0.05);与空白组相比,模型组血清中的IL-6含量升高(P<0.01),各治疗组小鼠血清中IL-6含量与模型组相比明显降低(P<0.01);与模型组相比,小鼠肿瘤组织中IL-6、JAK2、STAT3 mRNA及IL-6、p-JAK2、p-STAT3、c-Myc、Cyclin D1蛋白表达均显著降低,差异有统计学意义(P<0.01),其中联合用药高剂量组JAK2 mRNA表达降低,联合用药低剂量组Cyclin D1蛋白表达降低,差异有统计学意义(P<0.05)。结论:归芪白术方联合奥沙利铂可通过调节IL-6/JAK2/STAT3通路相关蛋白表达发挥抑瘤作用。
Objective:To explore the anti-tumor effect of Guiqibaizhu Recipe combined with oxaliplatin in IL-6/JAK2/STAT3 pathway on MFC gastric cancer tumor-bearing mice and its influence on inflammatory molecules.Methods:Established MFC gastric cancer tumor-bearing mouse models,randomly divided into model group,oxaliplatin group,oxaliplatin plus Guiqibaizhu Recipe high,medium and low dose groups,10 mice in each group,and another 10 healthy mice as a blank group;mice in each group were given corresponding drugs by oral gavage,blank group and model group were given normal saline for continuous treatment for 14 d.Mice were sacrificed and the spleen,thymus and tumor were weighed to calculate the organ index and tumor inhibition rate;HE staining to observe the pathological changes of mice tumor;ELISA method to detect content of IL-6 in mice serum;RT-qPCR and Western blotwere used to detect levels of IL-6,JAK2,STAT3 mRNA and IL-6,p-JAK2,p-STAT3 protein in tumor tissues;immunohistochemistry was used to detect c-Myc and Cyclin D 1 proteins in tumor tissues.Results:Tumor weight of mice in each treatment group were signifi⁃cantly lower than that in model group(P<0.01),weight of tumor in high and medium dose group of combined medication was signifi⁃cantly lower than that in oxaliplatin group(P<0.05,P<0.01);compared with blank group,spleen index and thymus index of mice in model group decreased(P<0.01);compared with model group,spleen index and thymus index of mice in oxaliplatin group decreased(P<0.01);compared with oxaliplatin group,there was a significant difference in spleen index in high dose groups of combined drug,and there was a significant difference in thymus index in high and medium dose groups of combined drug;compared with blank group,content of IL-6 in serum of model group increased(P<0.01),while content of IL-6 in serum of mice in each treatment group decreased significantly(P<0.01);compared with model group,expressions of IL-6,JAK2,STAT3 mRNA and IL-6,p-JAK2,p-STAT3,c-Myc,Cyclin D 1 protein in mice tumor tissue wer
