河南省洛阳市2016—2017年风疹病毒分子流行病学分析

Analysis on molecular epidemiology of rubella virus isolated from 2016 to 2017 in Luoyang city of China

摘要 目的分析河南省洛阳市2016—2017年风疹病例流行病学特征和风疹病毒分离株基因特征,为洛阳市风疹防控提供支持。方法搜集河南省洛阳市2016—2017年风疹病例流行病学信息,采用描述性流行病学的方法,分析其流行病学特征。采集病例咽拭子标本进行病毒分离,采用RT-PCR方法扩增风疹病毒E1基因片段,测序后获得序列,使用Mega7.0.26软件进行进化树分析、同源性分析、核苷酸特征和氨基酸特征分析。结果河南省洛阳市2016—2017年共报告病例232例,年龄集中于10~<15岁组和15~<20岁年龄组,发病高峰月份集中于3—6月。共分离到风疹病毒56株,均为2B基因型,核苷酸和氨基酸遗传距离分别为0~0.011和0~0.029。与2B基因参考株Rvi/Anqing.CHN/00/2B对比发现,氨基酸序列279位点异亮氨酸突变为缬氨酸,与Rvi/Beijing CHN/80/2A/Vac对比发现,氨基酸序列333位点丙氨酸突变为缬氨酸,这些突变与国内其他省份风疹分离株相同,E1蛋白基因糖基化位点以及抗原表位未发生变异。结论河南省洛阳市2016—2017年流行的风疹病毒2B基因型,核苷酸和氨基酸序列水平上高度保守,相关功能位点均未发生改变。 Objective To analyze the genetic characteristics of rubella virus isolated from 2016 to 2017 in Luoyang city of China. Methods Information of rubella cases reported in Luoyang from 2016 to 2017 was gathered. Samples of the cases were collected and virus isolation was performed.Fragments of E1 genes of the isolates were amplified by RT-PCR and the PCR products were sequenced.The phylogenetic tree was constructed and the gene characteristic analysis was performed by Mega7.0.26 software. Results A total of 232 rubella cases were reported from 2016 to 2017 in Luoyang city of China.Most cases were in 10—<15 years and 15—<20 years group and were reported from March to June.A total of 56 rubella virus strains were isolated, all of which belonged to genotype 2 B.The nucleotide acid and amino acid distance among them were 0—0.011 and 0—0.029, respectively.Comparing to Rvi/Anqing/CHN/00/2 B, all strains in Luoyang showed a mutation from Ile to Val at the 279 th amino acid;comparing to Rvi/Beijing/CHN/80/2 A/Vac, all Luoyang strains had a mutation from Ala to Val at the 333 rd amino acid that were found in strains from other provinces.There was no variation at known important functional sites. Conclusions The genotype of rubella virus isolated in Luoyang city of China during 2016 and 2017 is 2 B.All of the virus strains are highly conserved in nucleotide and amino acid sequences and there is no variation in known important functional sites.
