贝壳杉烯酸诱导烟草的系统获得性抗性防御番茄斑萎病毒侵染 被引量:1

AHK Inducing Systematic Acquired Resistance to Defense Tomato Spotted Wilt Orthotospovirus Infection in Nicotiana tabacum

摘要 【目的】番茄斑萎病毒(TSWV)在全球范围内广泛发生,危害多种作物引起严重的经济损失,筛选应用具有高效抑制活性的天然产物是防控TSWV引起的病害有效途径之一。前期研究发现三列叶蟛蜞菊(Wedelia trilobata)中提取的贝壳杉烯酸(AHK)具有抑制TSWV侵染的活性,本研究探明其具体的抑制机制。【方法】通过测定接种TSWV前、后用AHK处理的烟草植株相关防御酶活性以及RT-qPCR检测抗性相关主要代谢通路关键转录因子基因表达量,解析AHK防御TSWV侵染的机制。【结果】AHK在病毒接种前后处理中能诱导超氧化物歧化酶(SOD)活性升高而抑制TSWV的侵染;同时AHK能诱导茉莉酸(JA)通路重要转录因子MYC2、NAC表达量升高,增强了系统获得抗性。【结论】AHK主要通过诱导SOD酶活升高以及诱导JA通路增强寄主系统抗性从而抑制TSWV的侵染,具有作为植物源农药研发的潜力,为TSWV引起的病毒病绿色防控积累基础。 【Objective】Tomato spotted wilt orthotospovirus(TSWV)had occurred worldwide and caused seriously economic losses on many crops.It was one of the efficient path via screening of natural products with high inhibited activity to prevention and control of the diseases caused by TSWV.Our previous study indicated that kaurenic acid(AHK)extracted from Wedelia trilobata has a high effect against TSWV infection.In this study,we want to prove up the mechanism of AHK inhibition.【Method】Detected the activities of defense enzymes and the expression of systemic acquired resistance(SAR)-related genes by RT-qPCR from the tobacco plants before and post inoculated TSWV and treatment AHK,and analyzed the mechanism of inhibited by AHK.【Result】AHK induced superoxide dismutase(SOD)activity rised and inhibited TSWV infection.Meanwhile,AHK induced the increasing of MYC2 and NAC expression levels which were important transcription factors of Jasmonic Acid(JA)pathway.【Conclusion】AHK mainly induced the increasing of SOD activity and JA signaling pathway to enhance host SAR for inhibiting TSWV infection,and AHK had potential for the research and development of botanical pesticides for bases of prevention and contrl the diseases caused by TSWV.
