摘要
目的分析骨髓红系细胞的转录组数据,筛选出差异表达基因并对功能基因进行挖掘和分析,以期为红细胞成熟分化的分子机制提供理论依据。方法从TCGA数据库下载骨髓红系细胞转录组测序数据,对测序数据进行de novo拼接和功能注释;筛选出差异表达基因及在COG、GO、KEGG数据库中进行比对注释;基于Swiss-Prot功能注释结果,分析骨髓红系细胞分化成熟相关基因的功能及表达量变化。结果骨髓原红细胞和幼红细胞转录组样本中,共有5273个差异表达基因注释到Swiss-Prot数据库上,1993个差异表达基因具有详细的COG分类功能注释,5161个差异表达基因在GO数据库具有功能注释,2001个差异表达基因被注释到286条KEGG Pathway中。在筛选差异表达基因中,SDC2、LGALS2、MAFB、HMOX1在骨髓红系细胞中表达升高,ST6GAL2、CD34、FREM1、PKLR在骨髓红系细胞中表达降低。结论本研究提供了全面的骨髓红系细胞转录组信息,筛选出了一批在骨髓红系细胞分化成熟过程中具有重要功能的基因序列,如MAFB、HMOX1、PKLR、CD34等,为骨髓红系细胞分化成熟的调控机制研究提供了参考数据。
Objective To screen the differentially expressed genes to mine and analyze the functional genes by analyzing the transcriptomics data of bone marrow erythroid cells so as to provide a theoretical basis for the molecular mechanism of erythrocyte maturation and differentiation.Methods The transcriptome sequencing data of bone marrow erythroid cells were downloaded from TCGA database,and de novo concatenation and functional annotation were performed for the data.The differentially expressed genes were screened and compared in COG,GO and KEGG databases.Changes of function and expressions of genes related to differentiation and maturation of erythroid cells were analyzed based on the results of Swiss-Prot functional annotation.Results In transcriptome samples of bone marrow proerythroblasts and erythroblasts,a total of 5273 differentially expressed genes were annotated in Swiss-Prot database;1993 differentially expressed genes had detailed COG classification function annotation;5161 differentially expressed genes had function annotation in GO database,and 2001 differentially expressed genes were annotated in 286 KEGG pathways.In screening differentially expressed genes,expressions of SDC2,LGALS2,MAFB and HMOX1 were increased in bone marrow erythroid cells,while expressions of ST6GAL2,CD34,FREM1 and PKLR were decreased in bone marrow erythroid cells.Conclusion This study provides comprehensive transcriptome information of bone marrow erythroid cells,and it selects a number of gene sequences with important functions in process of differentiation and maturation of bone marrow erythroid cells such as MAFB,HMOX1,PKLR and CD34,which provides data reference for the regulatory mechanism of differentiation and maturation of bone marrow erythroid cells.
