摘要
目的探讨纳美芬对神经病理性疼痛大鼠的改善作用及可能机制。方法通过生物信息学技术分析IL-10在正常脑组织、胶质瘤表达情况以及不同级别胶质瘤表达情况;同时分析IL-10对胶质瘤病人预后影响。CCK-8方法检测IL-10对胶质瘤细胞系增殖能力影响。Transwell方法检测IL-10对胶质瘤侵袭能力影响。Western blot实验研究IL-10对基质金属蛋白酶(matrix metalloproteinase,MMP)-2表达影响。结果 IL-10在胶质瘤中高度表达,并且随着胶质瘤级别增高,IL-10表达量也增高;IL-10与胶质瘤病人较差的总生存期(overall survival,OS)和无病生存期(disease-free survival, DFS)相关。IL-10促进胶质瘤细胞增殖和侵袭,同时提高胶质瘤细胞MMP-2表达。结论 IL-10可能通过MMP-2通路促进胶质瘤增殖和侵袭。
Objective To investigate the effect of interleukin-10(IL-10) on the biological behavior of glioma.Methods We analyze the expression of IL-10 in normal brain tissues, different grades of glioma by bioinformatics technology. Then we determin the effect of IL-10 on the prognosis of glioma patients. The effect of IL-10 on proliferation and invasion was detected by CCK-8 assay and Transwell assay. The effect of IL-10 on matrix metalloproteinase(MMP)-2 expression was detected by Western blot. Results IL-10 was highly expressed in gliomas. IL-10 expression was associated with glioma grade. IL-10 high expression was associated with glioma patients′poor overall survival(OS) and poor disease-free survival(DFS). IL-10 promotes the proliferation and invasion of glioma cells, and the expression of MMP-2. Conclusion IL-10 could promotes proliferation and invasion of glioma cells by regulating MMP-2 pathway.
