摘要
目的探究舒芬太尼预处理对局灶性脑缺血-再灌注(Cerebral ischemia reperfusion,CIR)导致的大鼠脑损伤的保护作用及机制。方法30只雄性SD大鼠随机分为假手术组(Sham)、脑缺血-再灌注组(CIR)及舒芬太尼组(SUF),每组10只,线栓法建立大脑中动脉阻塞(MCAO)模型。Garcia法神经缺陷评分(NDS)检测神经功能;TTC染色检测脑梗死容积百分比(BIVP);HE染色与Nissl染色观察脑组织结构变化;湿法消解法检测大鼠脑组织总钙含量;Western blot检测大鼠海马区SYN、BDNF及p-CREB蛋白表达。结果与CIR组相比,舒芬太尼预处理显著改善大鼠的神经功能,显著降低大鼠的脑梗死容积,改善脑组织结构并增加海马区神经元数量,显著降低大鼠脑内总钙含量,并显著增加大鼠海马区SYN、BDNF及p-CREB蛋白表达。结论舒芬太尼对大鼠脑缺血-再灌注所致脑损伤具有一定的神经保护作用,其作用机制可能与激活脑内BDNF-CREB信号通路有关。
Objective To investigate the protective effect of sufentanil preconditioning on cerebral ischemia reperfusion(CIR)induced brain injury in rats and its mechanism.Methods Thirty male SD rats were randomly divided into Sham group(SHAM),cerebral ischemia-reperfusion group(CIR)and sufentanil group(SUF),with 10 rats in each group.Middle cerebral artery occlusion(MCAO)model was established by suture embolization.The neural function of rats was measured by Garcia neural deficiency score(NDS).TTC staining was used to detect the percentage of cerebral infarction volume(BIVP).HE staining and Nissl staining were used to observe the changes of brain tissue structure.The content of total calcium in brain tissue of rats was determined by wet digestion method.The protein expressions of Syn,BDNF and p-CREB in rat hippocampus were detected by Western blot.Results Compared with the CIR group,sufentanil preconditioning significantly improved the neurological function of rats,significantly reduced the brain infarct volume,improved brain tissue structure and increased the number of neurons in hippocampus,significantly decreased the content of total calcium in rat brain,and significantly increased the proteinsexpression of SYN,BDNF and p-CREB in hippocampus of rats.Conclusion Sufentanil has a neuroprotective effect on brain injury caused by cerebral ischemia-reperfusion in rats,and its mechanism may be related to the activation of BDNF-CREB signaling pathway in the brain.
