摘要
目的:探索针刀、电针治疗对早期膝骨关节炎(KOA)兔模型关节软骨细胞结构及Ⅱ型胶原的表达情况。方法:30只新西兰兔按体质量随机分为空白组(6只)及KOA造模组(24只),KOA造模组应用改良Videman法制动4周建立早期KOA兔模型。干预结束后,对兔股骨内侧髁软骨进行免疫荧光双标染色(激光共聚焦显微镜观察),应用ELISA法检测兔血清、关节液CTX-2的含量。结果:①治疗后针刀组、电针组血清(P=0.091,P=0.657)、关节液(P=0.066,P=0.032)CTX-2含量降低。②治疗后针刀组、电针组细胞核(P=0.058,P=0.051)、F-actin骨架蛋白(P=0.184,P=0.220)和Ⅱ型胶原(P=0.168,P=0.236)IOD表达降低,但差异无统计学意义。结论:针刀、电针对早期KOA兔模型膝关节软骨细胞及Ⅱ型胶原蛋白代谢具有一定保护作用,但效果不如塞来昔布。
Objective:To explore the effects of acupotomy and electro-acupuncture on structure of articular chondrocyte and expression of typeⅡcollagen in rabbit model of early knee osteoarthritis(KOA).Methods:30 New Zealand rabbits were randomly divided into the blank group(n=6)and the model group(n=24).The KOA model was established by modified Videman method(immobilization for 4 weeks).After the interventions of acupotomy and electro-acupuncture,the rabbit femoral condyle cartilage was stained with double immunofluorescence and observed by laser confocal microscopy.The contents of CTX-2 in serum and in synovial fluid were detected by ELISA.Results:After the interventions,the contents of CTX-2 in serum(P=0.091,P=0.657)and in synovial fluid(P=0.066,P=0.032)were decreased in the acupotomy group and in the electro-acupuncture group.The IOD expressions of the nucleus(P=0.058,P=0.051),F-actin(P=0.184,P=0.220)and typeⅡcollagen(P=0.168,P=0.236)were decreased in the acupotomy group and in the electro-acupuncture group,but there were no statistical differences.Conclusion:Acupotomy and electro-acupuncture have certain protective effects on the articular chondrocytes and the metabolism of typeⅡcollagen in rabbits with early KOA,however the effects are not as effective as Celecoxib.
