感染后肠易激综合征小鼠肠道黏膜屏障功能的动态改变 被引量:4

Dynamic changes of intestinal mucosal barrier function in mice with post-infectious irritable bowel syndrome

摘要 [目的]探讨感染后肠易激综合征对小鼠肠黏膜屏障功能的影响。[方法]选择8周龄成年雄性NIH小鼠40只,随机分为对照组和感染2、4、8周组,每组10只。采用旋毛虫感染法制作感染后肠易激综合征模型。于感染后2、4、8周,HE染色观察肠道病理改变、记录小鼠体质量、大便含水量、肠道动力情况。采用改良分光光度法检测血清D-乳酸,采用酶联免疫吸附试验检测分泌性免疫球蛋白(sIgA)含量变化。[结果]感染后2周组小鼠肠道出现急性炎症反应,体质量显著降低(P<0.05),感染后2周组体质量基本恢复至正常。各感染组小鼠排便时间快于对照组,且感染后8周组小鼠的排便时间最快,各感染后8周组恢复接近正常。各感染组小鼠粪便含水百分数均高于对照组(P<0.05),且以感染后2周组粪便含水量最多。与对照组比较,感染后2周组小鼠D-乳酸水平和sIgA水平明显升高(P<0.05);感染后8周组小鼠D-乳酸水平和sIgA水平接近正常。[结论]D-乳酸含量及sIgA水平升高可能是导致感染后肠易激综合征小鼠肠黏膜通透性增加以及粪便含水百分数增高的原因之一,参与了感染后肠易激综合征的发生机制。 [Objective]To investigate the change of intestinal mucosal barrier function in mice with post-infectious irritable bowel syndrome.[Methods]The Forty 8-week-old NIH male mice were randomly divided into the control group and infection groups,with 20 mice in each group.The model of irritable bowel syndrome was established by Trichinella spiralis infection.Pathological changes of gut were observed by HE staining,the changes in body weight,fecal water content,intestinal motility,D-lactic acid levels and Secretory immunoglobulin(SIgA)of mice in the control group and infection groups(2 weeks,4 weeks and 8 week after trichinella infection)were compared.[Results]After 2 weeks infection,acute inflammation of the intestinal tract was observed and the body weight of mice were significantly decreased(P<0.05).Eight weeks after infection,body weight of mice recovered to normal.Compared with control group,the defecation time in infection groups was significantly decreased 2 weeks after the infection and recovered to normal 8 weeks after the infection.Fecal water content in infection groups was significantly higher than that in the control group 2 weeks after the infection and recovered to normal 8 weeks after the infection.The level of D-lactic acid and SIgA 2 weeks after the infection were significantly increased in the infection group.[Conclusion]The level of D-lactic acid and SIgA show effect on the defecation time and fecal water content of the mice with post-infectious irritable bowel syndrome,which is the possible mechanisms for post-infectious irritable bowel syndrome.
