摘要
目的探讨多聚ADP核糖聚合酶-1(poly(ADP-ribose)polymerase-1,PARP-1)与程序性死亡蛋白(programmed cell death protein-1,PD-1)、程序性死亡蛋白配体-1(programmed cell death protein ligand-1,PD-L1)在子宫内膜癌组织和正常子宫组织中的表达水平及其临床意义。方法收集同济大学附属东方医院诊治的74例子宫内膜癌组织和55例正常子宫内膜组织及临床病理资料,采用免疫组织化学法检测各组织PARP-1与PD-1、PD-L1的表达水平,分析子宫内膜癌组织中PARP-1与PD-1、PD-L1表达水平、三者的相关性及与临床意义。结果PARP-1与PD1在子宫内膜癌和正常子宫内膜组织中表达差异有统计学意义(P均<0.05)。在子宫内膜癌组织中,PARP-1阳性表达与浸润深度、组织分化程度及临床分期相关(P均<0.05),与年龄、病灶大小及淋巴结转移无关。PD-1蛋白的表达与病灶大小、肌层浸润深度、组织分化程度及临床分期显著相关(P均<0.05),与年龄、淋巴结转移无关。PD-L1的表达与组织分化程度及临床分期相关(P均<0.05),与年龄、病灶大小、肌层浸润深度及淋巴结转移均无关。子宫内膜癌组织PARP-1与PD-1、PD-L1表达呈负相关性(r值分别为0.245及0.315,P均<0.05)。结论PARP-1、PD-1、PD-L1在子宫内膜癌组织中表达升高,与组织分化程度、浸润深度及临床分期等临床因素相关,有望作为子宫内膜癌早期诊断、预测复发和评价预后的指标。
Objective To explore the expression of poly(ADP-ribose)polymerase-1(PARP-1),programmed cell death protein-1(PD-1),programmed cell death protein ligand-1(PD-L1)in endometrial cancer and its clinical significance.Methods The clinical and pathological data of 74 cases of endometrial cancer tissues and 55 cases of normal endometrial tissues were collected,and the expression levels of PARP-1,PD-1 and PD-L1 were detected by immunohistochemical method.The expression level,correlation and clinical significance of PARP-1,PD-1 and PD-L1 in endometrial cancer tissues were analyzed.Results There was a significant difference in the expression of PARP-1 and PD1 between endometrial cancer and normal endometrial tissues(both P<0.05).In endometrial cancer tissues,the positive expression of PARP-1 was related to the depth of invasion,degree of tissue differentiation and clinical stage(all P<0.05),but not related to age,lesion size and lymph node metastasis.The expression of PD-1 protein was significantly related to the size of the lesion,the depth of myometrial infiltration,the degree of tissue differentiation,and the clinical stage(all P<0.05),and was not related to age and lymph node metastasis.The expression of PD-L1 was related to the degree of tissue differentiation and clinical stage(all P<0.05),but was not related to age,lesion size,depth of myometrial invasion,and lymph node metastasis.The expression of PARP-1 in endometrial cancer tissue was negatively correlated with PD-1 and PD-L1(r values were 0.245 and 0.315,respectively,P<0.05).Conclusion PARP-1,PD-1,and PD-L1 are elevated in endometrial cancer tissues,which are of great significance to the degree of tissue differentiation,the depth of invasion and clinical stage.They are expected to be used as an indicator for early diagnosis,prediction of recurrence and evaluation of prognosis of endometrial cancer.
