桂千金子提取物对金黄色葡萄球菌体外抗菌作用及机制的初步研究 被引量:2

摘要 目的:探讨桂千金子提取物对金黄色葡萄球菌的体外抗菌活性及作用机制。方法:通过体外抑菌试验,测定桂千金子乙醇提取物及其乙酸乙酯、正丁醇萃取物对金黄色葡萄球菌的最低抑菌浓度(MIC)和最低杀菌浓度(MBC),比较标准菌株金黄色葡萄球菌(SA)及临床分离的金黄色葡萄球菌(MSSA)和耐甲氧西林金黄色葡萄球菌(MRSA)的抗菌活性大小。测定桂千金子乙醇提取物作用后MRSA碱性磷酸酶(AKP)活性、外流DNA/RNA大分子含量、胞内蛋白含量的变化,采用电镜观察MRSA细胞形态。结果:桂千金子提取物对3种供试菌株的抑菌圈直径均大于15 mm,MIC为1.56~12.50 mg/mL,MBC为3.13~25.00 mg/mL,抗菌强度排序为乙醇提取物≈乙酸乙酯萃取物>正丁醇萃取物;与空白对照组比较,实验组AKP活性升高(P<0.05),各实验组与空白组外流DNA/RNA大分子含量和胞内蛋白含量比较,差异均无统计学意义(均P>0.05);电镜下MRSA菌体细胞变形,表面粗糙有颗粒附着。结论:桂千金子提取物对金黄色葡萄球菌及其耐药菌株均具有显著的抗菌活性,其抗菌作用可能与细胞壁结构破损致通透性发生改变有关。 Objective:To study antibacterial activity and mechanism of extracts from Polygonum runcinatum Buch.-Ham.against Staphylococcus aureus.Methods:Minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of the ethanol extract,ethyl acetate extract and n-butanol extract of Polygonum runcinatum Buch.-Ham.on MRSA were determined by in vitro antibacterial test.The antibacterial activities of Staphylococcus aureus standard strain(SA),clinically isolated Staphylococcus aureus(MSSA)and methicillin resistant Staphylococcus aureus(MRSA)were compared.The alkaline phosphatase(AKP)activity,the contents of outflow DNA/RNA macromolecules and intracellular protein were detected.The morphology of MRSA cells was observed by electron microscopy.Results:The inhibitory circle diameter of extracts to the tested strains was greater than 15 mm,MIC was 1.56-12.50 mg/mL,and MBC was 3.13-25.00 mg/mL.The order of antibacterial strength was ethanol extract≈ethyl acetate extract>n-butanol extract.Compared with the blank group,the AKP activity was increased in the experimental group(P<0.05),and there was no significant difference in the contents of outflow DNA/RNA macromolecules and intracellular protein(P>0.05).MRSA cells were deformed,with rough surface and particle attachment.Conclusion:The extracts of Polygonum runcinatum Buch.-Ham.had significant antibacterial activity against Staphylococcus aureus and its drug-resistant strains.Its antibacterial effect may be related to the change of permeability caused by the damage of cell wall struc-ture.
