摘要
目的:探讨金佛手醇提液对哮喘小鼠肺组织病理变化和基质金属蛋白酶-9(MMP-9)水平的影响。方法:选择BALB/c雌性小鼠50只分为正常组、模型组、金佛手醇提液高(30 g/kg)、低剂量组(15 g/kg)、阳性对照组(地塞米松,5 mg/kg),每组各10只。哮喘小鼠模型采用卵白蛋白激发诱导。将建模成功后的小鼠干预4周后,处死小鼠,光镜观察病理学变化,ELISA检测肺组织MMP-9水平。结果:与正常组相比,模型组小鼠肺组织内有大量炎性细胞,嗜酸性粒细胞、中性粒细胞和淋巴细胞浸润;金佛手低剂量组、高剂量组和阳性药物对照组肺组织炎性细胞浸润与模型组相比明显减少。模型组肺组织MMP-9水平明显高于正常组,金佛手低剂量组、高剂量组及阳性药物对照组MMP-9水平显著低于模型组(P <0.05)。金佛手低剂量组、高剂量组与阳性药物组组间比较无明显差异(P> 0.05)。结论:金佛手醇提液可减轻哮喘小鼠病理改变,降低肺组织MMP-9的水平。
Objective:To investigate the effects of alcohol extract of Bergamot on pathological changes and MMP-9 levels in lung tissue of asthmatic mice.Methods:The mouse model of asthma was stimulated by ovalbumin.Mice were divided into normal group,model group,alcohol extract of Bergamot high( 30 g/kg)group,low dose group( 15 g/kg) and positive drug control group( Dexamethasone,5 mg/kg),with 10 mice in each group.After 4 weeks of intervention,the mice were sacrificed for pathological observation by optical microscope,and the content of MMP-9 was detected by enzyme-linked immunosorbent assay( ELISA).Results:After successful modeling,asthma-like symptoms appeared in mice.Compared with the normal group,a large number of inflammatory cells,eosinophils,neutrophils and lymphocytes were observed in the lung tissues of the model group,while the infiltration of inflammatory cells in the lung tissues of the alcohol extract of Bergamot low dose.MMP-9 level in the alcohol extract of Bergamot low dose group,high dose group and positive drug control group was lower than that of model group,MMP-9 level in the model group was higher than normal group,while( P < 0.05).Conclusion:Alcohol extract of Bergamot can reduce the pathological changes of lung tissue in asthmatic mice,lower the MMP-9 level.
