摘要
目的:分析纯镁微弧氧化-壳聚糖/海藻酸钠载铜膜层的细胞相容性及抑菌性。方法:以纯度为99.9%的镁片制作20片试件,分为4组各5片,甲组表面为纯镁微弧氧化层,乙组为纯镁微弧氧化-壳聚糖复合膜层,丙组为纯镁微弧氧化-壳聚糖/海藻酸钠膜层,丁组为纯镁微弧氧化-壳聚糖/海藻酸钠载铜膜层,比较四组试件的显微形态、湿润角、细胞相容性及抑菌性。结果:试件表面微孔、裂纹数量,甲组>乙组>丙组>丁组;细胞铺展面积,丁组>丙组>乙组>甲组;各组试件的湿润角比较,差异无统计学意义(P>0.05);在大肠埃希菌培养液中培养24 h后,菌落数甲组>乙组>丙组>丁组,抑菌性丁组>丙组>乙组,差异均有统计学意义(P<0.05)。结论:纯镁微弧氧化-壳聚糖/海藻酸钠载铜膜层可有效封闭镁片表面微孔,且细胞相容性、抑菌性良好,效果优于纯镁微弧氧化层、纯镁微弧氧化-壳聚糖复合膜层及纯镁微弧氧化-壳聚糖/海藻酸钠膜层。
Objective:To analyze cell compatibility and antibacterial activity of micro-arc oxidized pure Magnesium chitosan/sodium alginate containing copper composite film.Methods:20 test pieces were made of magnesium flakes with a purity of 99.9%and then were divided into 4 groups with 5 pieces each.The surface of group A was micro-arc oxidized pure Magnesium layer;the surface of group B was micro-arc oxidized pure Magnesium-chitosan composite film layer;the surface of group C was micro-arc oxidized pure Magnesium-chitosan/sodium alginate film layer;and the surface of group was micro-arc oxidized pure Magnesium chitosan/sodium alginate containing copper composite film layer.The microscopic morphology,the wetting angle,the cell compatibility and the antibacterial activity were compared among the four groups.Results:The number of micro-pores and cracks on the surface of the specimen:group A>group B>group C>group D;the cell spreading area:group D>group C>group B>group A.There was no significant difference in the wetting angle among the four groups(P>0.05).After culturing in Escherichia coli culture medium for 24 hours,the number of colonies was as follows:group A>group B>group C>group D;the antibacterial activity was as follows:group D>group C>group B;and the differences were statistically significant(P<0.05).Conclusion:Micro-arc oxidized pure Magnesium chitosan/sodium alginate containing copper composite film can effectively seal the micropores on the surface of the Magnesium sheets,and has good cell compatibility and antibacterial properties.Moreover,it is superior to micro-arc oxidized pure Magnesium layer,micro-arc oxidized pure Magnesium-chitosan composite film layer and micro-arc oxidized pure Magnesium-chitosan/sodium alginate film layer.
