HPLC法检测蔓荆子中蔓荆子黄素的含量

摘要 目的:建立蔓荆子中蔓荆子黄素含量的测定方法。方法:采用高效液相色谱法,选择Agilent SB C18柱(250 mm×4.6 mm,5μm)为色谱柱,以乙腈和0.1%磷酸溶液为流动相(梯度洗脱),流速1.0 mL/min,检测波长254 nm。结果:蔓荆子黄素的线性关系方程为Y=1.42×10^(4)X-2.172×10^(2),在12.5~200μg/mL范围内的峰面积与进样浓度线性关系良好(R^(2)=0.9997)。精密度、重复性及稳定性试验的相对标准偏差(RSD)分别为0.71%、1.05%、0.96%,平均加样回收率为98.19%,其RSD为1.15%。蔓荆子中蔓荆子黄素的平均含量为0.0554%。结论:建立的高效液相色谱法方法简便、准确、快速,重复性好,专属性好,可应用于蔓荆子中蔓荆子黄素含量测定。 Objective:To establish a method for the determination of vitexin in Vitex trifolia.Methods:Agilent SB C18 column(250 mm×4.6 mm,5μm)was used as the chromatographic column with acetonitrile and 0.1%phosphoric acid as mobile phases(gradient elution)at a flow rate of 1.0 mL/min.The detection wavelength was 254 nm.Results:The linear equation of Vitexin was Y=1.42×10^(4)X-2.172×10^(2).The peak area in the range of 12.5-200μg/mL had a good linear relationship with the injection concentration(R^(2)=0.9997).The relative standard deviations(RSD)of precision,repeatability and stability tests were 0.71%,1.05%and 0.96%,respectively.The average recoveries were 98.19%and RSD was 1.15%.The average content of Vitexin in Vitex trifolia was 0.0554%.Conclusion:The established HPLC method is simple,accurate,rapid,reproducible and specific.It can be applied to the determination of Vitexin in Vitex trifolia.
