摘要
【目的】为分析USP9Y与ND1基因在玉树牦牛种内的遗传变异,并对玉树牦牛种群优化工作给予分子水平上的理论依据和科学建议。【方法】研究对100头玉树牦牛(40♂,60♀)目标基因片段(USP9Y与ND1基因)进行PCR扩增测序和多态性检测,并基于实验结果为玉树牦牛育种提供建议。【结果】玉树牦牛USP9Y核苷酸序列长度为210 bp,ND1核苷酸序列长度为220 bp,未在玉树牦牛群体内发现突变位点,得出玉树牦牛群体中USP9Y与ND1基因的杂合度低,表明玉树牦牛群体封闭,有近交衰退的趋势。通过与青海范围内牦牛ND1基因序列(高原、雪多、环湖、阿什旦、大通和野牦牛)比对,得出玉树牦牛与高原和雪多牦牛距离较近,而与阿什旦、环湖、大通和野牦牛遗传距离较远。建议在培育玉树牦牛核心群时,应划区进行,应用优势种如环湖公牦牛或其他遗传距离较远公牦牛的冻精配种及适度引种。【结论】玉树牦牛群体内USP9Y与ND1基因部分片段无基因多态性,表示以上玉树牦牛群体杂合度低,建议科学配种并合理引进优势畜种进行培育,提高玉树牦牛品种的遗传潜力。
[Objective]To analyze the genetic variation of USP9 Y and ND1 genes in Yushu yak,and give theoretical basis and scientific suggestions on Yushu yak breeding at the molecular level.[Method]In this study,PCR amplification,sequencing and polymorphism detection of target gene fragments(USP9 Y and ND1 genes)in 100 Yushu yaks(40♂,60♀)were conducted,and breeding suggestions for Yushu yak varieties(populations)were put forward based on the experimental results.[Result]The length of USP9 Y nucleotide sequence of Yushu yak was 210 bp,while the length of ND1 nucleotide sequence of Yushu yak was 220 bp.However,no mutation site was found in Yushu yak population.It is concluded that the heterozygosity of USP9 Y and ND1 genes in Yushu yak population is low,indicating that Yushu yak population is closed and has the effect of inbreeding decline.The ND1 gene sequences of Yaks(Plateau,Xueduo,Huanhu,Ashidan,Datong and Wild Yak)were downloaded from Qinghai Province.The results showed that Yushu yak was closer to Plateau and Xueduo yak,but farther to Ashidan,Huanhu,Datong and Wild yak.It is suggested that when cultivating Yushu yak core group,dominant male yak frozen semen breeding and moderate introduction should be carried out on zone location.[Conclusion]There is no gene polymorphism in some fragments of USP9 Y and ND1 genes in Yushu yak population,indicating that the heterozygosity of the above two genes is low in Yushu yak population.Scientific breeding and introduction of dominant breeds are suggested for cultivation so as to increase the genetic potential of Yushu yak varieties.
