右美托咪定对老年小鼠海马和杏仁核半胱氨酸蛋白酶-3基因表达影响的实验研究 被引量:3

摘要 目的:探讨右美托咪定对老年小鼠海马和杏仁核半胱氨酸蛋白酶-3(Caspase-3)基因表达的影响。方法:48只老年(18月龄)小鼠随机平分为三组:对照组、舒芬太尼组与右美托咪定组。舒芬太尼组与右美托咪定组分别给予5μg/kg舒芬太尼与5μg/kg右美托咪定进行腹腔注射,对照组给予等容量0.9%氯化钠溶液腹腔注射,每周1次,持续8周,检测Caspase-3表达变化情况。结果:舒芬太尼组与右美托咪定组干预第4周与第8周的逃避潜伏期低于对照组(均P<0.05),穿越平台次数与处于平台所在象限时间高于对照组(P<0.05),舒芬太尼组与右美托咪定组对比差异有统计学意义(P<0.05)。舒芬太尼组与右美托咪定组干预第4周与第8周的血清去甲肾上腺素含量高于对照组(均P<0.05),TNF-α含量低于对照组(P<0.05),舒芬太尼组与右美托咪定组对比差异有统计学意义(P<0.05)。舒芬太尼组与右美托咪定组干预第8周的海马和杏仁核Caspase-3基因、蛋白表达水平低于对照组(均P<0.05),舒芬太尼组低于右美托咪定组(P<0.05)。结论:右美托咪定在老年小鼠的应用能抑制海马和杏仁核Caspase-3基因与蛋白表达,也可抑制肿瘤坏死因子-α(TNF-α)的释放,提高血清去甲肾上腺素含量,从而有利于小鼠的认知功能恢复。 Objective:To investigate the effect of dexmedetomidine on Caspase-3 gene expression in hippocampus and amygdala of aged mice.Methods:48 cases of elderly(18-month-old)mice were randomly divided into control group,sufentanil group,dexmedetomidine group.The sufentanil group and dexmedetomidine group were injected with 5μg/kg sufentanil and 5μg/kg dexmedetomidine intraperitoneally respectively,and the control group was injected intraperitoneally with equal volume of 0.9%sodium chloride solution,once a week for 8 weeks.The expression of Caspase-3 was detected.Results:The escape latency of the sufentanil group and the dexmedetomidine group at the 4th and 8th week of the intervention was lower than that of the control group,and the times of crossing the platform and the time in the quadrant of the platform were higher than those of the control group(all P<0.05).The content of serum norepinephrine in sufentanil group and dexmedetomidine group at the 4th and 8th week of intervention was higher than that in the control group,and the content of TNF-αwas lower than that in the control group(all P<0.05).The expression levels of Caspase-3 gene and protein in hippocampus and amygdala in sufentanil group and dexmedetomidine group were lower than those in control group at the 8th week of intervention,and that in sufentanil group was lower than that in dexmedetomidine group(all P<0.05).Conclusion:The application of dexmedetomidine in aged mice can inhibit the expression of Caspase-3 gene and protein in the hippocampus and amygdala,as well as the release of TNF-α,and increase the serum norepinephrine content,which is beneficial to cognitive function recovery in mice.
