摘要
目的基于TLR4/NF-κB信号通路探讨银莱汤治疗幼龄大鼠肺炎的作用机制。方法将40只SPF级4周龄雄性大鼠随机分为4组,分别为正常组、模型组、银莱汤组和醋酸泼尼松组,每组10只。使用LPS(0.5 mg/mL)连续雾化吸入3 d制备肺炎模型。第4天麻醉后取血及相关脏器组织,采用ELISA试剂盒检测各组大鼠血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)水平的变化;HE染色观察肺组织形态学变化;Western blotting法检测肺组织中TLR4、My D88、TRAF6、NF-κB p65、p-IκBα、IκBα蛋白的表达。结果与正常组相比,模型组大鼠的血清炎症因子TNF-α、IL-6、IL-1β含量明显升高(P<0.05),肺组织中的TLR4、MyD88、TRAF6、p-IKBα、NF-κB p65蛋白含量表达明显升高(P<0.05),IKBα蛋白含量表达明显降低(P<0.05)。与模型组相比,银莱汤组和醋酸泼尼松组大鼠的血清炎症因子TNF-α、IL-6、IL-1β含量明显降低(P<0.05),肺组织中的TLR4、MyD88、TRAF6、p-IKBα、NF-κB p65蛋白含量表达明显降低(P<0.05),IKBα蛋白含量表达明显升高(P<0.05)。结论银莱汤可以通过调控TLR4/NF-κB炎症信号通路中的TLR4、MyD88、TRAF6、IKBα、p-IKBα、NF-κB p65蛋白表达水平,降低血清中炎症因子TNF-α、IL-6、IL-1β水平。
Objective:To explore the mechanism of action of Yinlai Decoction in the treatment of pediatric pneumonia based on TLR4/NF-κB signaling pathway.Methods:Forty SPF-rated 4-week-old male rats were randomly divided into four groups,namely,normal group,model group,Yinlai Decoction group and prednisone acetate group,with 10 rats in each group.Pneumonia models were prepared using LPS(0.5 mg/mL)by continuous nebulized inhalation for 3 days.Blood and related organ tissues were taken after anesthesia on the 4 th day.Changes in the levels of inflammatory factors TNF-α,IL-6 and IL-1βin the serum of rats in each group were detected by ELISA kits;HE staining was used to observe morphological changes in lung tissue;detection of TLR4,MyD88,TRAF6,NF-κB p65,p-IκBα,IκBαprotein expression in lung tissue was conducted by Western blotting.Results:Compared with the normal group,the serum inflammatory factors TNF-α,IL-6 and IL-1βlevels were significantly higher in the model group(P<0.05);the expression of TLR4,MyD88,TRAF6,p-IKBα,NF-κB p65 protein content was significantly higher in lung tissue(P<0.05);IKBαprotein content expression was significantly reduced(P<0.05).Compared with the model group,the serum inflammatory factors TNF-α,IL-6 and IL-1βlevels were significantly lower in rats in the Yinlai Decoction and prednisone acetate groups(P<0.05);the expression of TLR4,MyD88,TRAF6,p-IKBα,NF-κB p65 protein content in lung tissue was significantly reduced(P<0.05);the expression of IKBαprotein content was significantly higher(P<0.05).Conclusion:Yinlai Decoction can regulate TLR4,MyD88,TRAF6,IKBα,p-IKBα,NF-κB p65 protein expression levels in TLR4/NF-κB inflammatory signaling pathway and reduce serum levels of inflammatory factors TNF-α,IL-6,IL-1β.
