摘要
目的通过癌症基因组图谱(TCGA)数据库评估SLC47A1基因在肾透明细胞癌(ccRCC)的表达情况及临床应用价值。方法使用Wilcoxon符号秩和检验分析SLC47A1 mRNA在ccRCC组织和癌旁组织中的表达差异。采用RT-qPCR检测7例ccRCC患者中SLC47A1的相对表达。用Wilcoxon符号秩和检验和Logistic回归分析临床病理特征与SLC47A1之间的关系。使用Cox回归和Kaplan-Meier(KM)方法以探讨SLC47A1与ccRCC患者临床病理特征及预后之间的关联。应用基因集富集分析(GSEA)软件探索SLC47A1调控ccRCC进展潜在的信号通路。结果 SLC47A1 m RNA在ccRCC组织的表达量明显高于其在癌旁组织中的表达,差异有统计学意义(P<0.05)。SLC47A1在ccRCC中的低表达与肿瘤细胞低分化(OR=0.29,95%CI:0.16~0.49),临床高分期(OR=0.37,95%CI:0.22~0.62),发生远处转移(OR=0.44,95%CI:0.26~0.73)等密切相关(P均<0.05)。KM生存分析表明,SLC47A1低表达患者的预后相较于SLC47A1高表达的患者欠佳,差异有统计学意义(P<0.05)。多因素Cox分析显示SLC47A1低表达量可以作为ccRCC独立预后预测因素,且与总体生存率呈负相关(HR=0.76,95%CI:0.67~0.85;P=4.92E-06)。结论 SLC47A1低表达可能是ccRCC不良预后的潜在生物学标志物。
Objective To evaluate the expression of SLC47A1 gene in clear cell renal cell carcinoma(ccRCC)and its clinical application value through the cancer genome atlas(TCGA)database.Method Wilcoxon signed rank sum test was used to analyze the expression differences of SLC47A1 mRNA in ccRCC tissues and adjacent tissues.RT-qPCR was used to detect the relative expression of SLC47A1 in 7 patients with ccRCC.The relationship between clinicopathological characteristics and SLC47A1 was analyzed by Wilcoxon signed rank test and Logistic regression.Cox regression and KaplanMeier(KM)methods were used to explore the association between SLC47A1 and the clinicopathological characteristics and prognosis of ccRCC patients.Gene Set Enrichment Analysis(GSEA)software was used to explore the potential signal pathways of SLC47A1 regulating the progress of ccRCC.Result The expression of SLC47A1 mRNA level in ccRCC tissue was significantly higher than that in paracancerous tissues(P<0.05).The low expression level of SLC47A1 in cc RCC was closely related to the poor differentiation of tumor cells(OR=0.29,95%CI:0.16-0.49),high clinical stage(OR=0.37,95%CI:0.22-0.62),and distant metastasis(OR=0.44,95%CI:0.26-0.73)(all P<0.05).KM survival analysis showed that the prognosis of patients with low SLC47A1 expression level was poorer than that of patients with high SLC47A1 expression level(P<0.05).Multivariate Cox analysis showed that low expression level of SLC47A1 could be used as an independent prognostic predictor of ccRCC,and it was negatively correlated with overall survival(HR=0.76,95%CI:0.67-0.85;P=4.92 E-06).Conclusion The low expression level of SLC47A1 might be a potential biomarker for poor prognosis of cc RCC.
