摘要 【目的】为了对枣GH3基因家族进行全面分析并对家族成员的功能进行研究。【方法】本研究对枣GH3基因家族进行鉴定并对其理化性质、基因结构、启动子元件、表达模式等进行了系统分析。【结果】基于枣基因组数据共鉴定得到19个枣GH3基因家族成员,每个家族成员均包括了GH3蛋白保守结构域,编码氨基酸长度在149~629 aa。等电点在5.21~8.90,不稳定系数范围在34.34~56.54,大部分成员为酸性的稳定蛋白均被定位在细胞质上。二级结构表明α-螺旋和无规则卷曲占比最高。系统进化树分析将19个枣GH3基因分成了3大类,每个类别中所包括的枣GH3基因家族成员数量差别不大。通过分析基因信息和蛋白特征发现motif3和motif6保守度最高,基因结构都较为简单,包括2到5个外显子且均有UTR组成。启动子元件分析表明,枣GH3基因可能受光照、茉莉酸甲酯和无氧诱导等多种因素的调控。染色体定位表明,19个ZjGH3基因中有4个(ZjGH3-16、ZjGH3-17、ZjGH3-18和ZjGH3-19)未被定位到枣的12条染色体上,其他的15个家族成员被定位到枣的Chr4、Chr8、Chr9、Chr10、Chr11和Chr12染色体上,同时分析到有8对基因存在基因复制事件。通过分析ZjGH3基因在‘壶瓶枣’果实发育期的表达情况表明,可将19个ZjGH3基因分成3大类,分别为在H1(白熟期)表达量最高、H2(半红期)表达量最高和H3(全红期)表达量最高。【结论】本研究共鉴定出19个枣GH3基因,其中ZjGH3-16和ZjGH3-17在果实发育中起重要作用。 【Objective】In order to analyze the GH3 gene family and study the function of family members in Ziziphus jujuba.【Method】In this study,the GH3 gene family was identified and its physical and chemical properties,gene structure,promoter elements and expression pattern were systematically analyzed.【Result】The 19 ZjGH3 gene family members were identified based on the genomic data.Each member included a conserved domain of GH3 protein,coding for amino acid length of 149-629 aa.The pI ranged from 5.21 to 8.90,and the instability index ranged from 34.34 to 56.54.Most of them are acidic stable proteins and localized in cytoplasmic.The secondary structure showed that the alpha helix and random coil have the highest proportion.Based on the phylogenetic analysis 19 Z.jujuba GH3 genes were divided into 3 groups,and the number of GH3 family genes included in each group had a little difference.According to the analysis of gene information and protein characteristics,motif3 and motif6 were found to be the most conserved genes with simple gene structure,including 25 exons and UTR.Promoter analysis indicated that the ZjGH3 genes might be regulated by light,MeJA and anaerobic induction.Chromosome mapping showed that 4 of the 19 ZjGH3 genes(ZjGH3-16,ZjGH3-17,ZjGH3-18 and ZjGH3-19)were not located on 12 chromosomes of Z.jujuba,and the other 15 family members were located on Chr4,Chr8,Chr9,Chr10,Chr11 and Chr12,at the same time,8 pairs of genes were found to have gene duplication events.By analyzing the expression of ZjGH3 gene in fruit development stage of Hupingzao,the results showed that 19 ZjGH3 genes could be divided into three groups,the highest expression of ZjGH3 gene in H1(white ripening period),the highest expression of H2(half red period)and the highest expression of H3(full red period),respectively.【Conclusion】A total of 19 ZjGH3 genes were identified,ZjGH3-16 and ZjGH3-17 played an important role during fruit development.
