薯蓣皂苷元下调EZH2对结直肠癌HCT116细胞增殖和凋亡的影响 被引量:3

摘要 目的:研究薯蓣皂苷元对结直肠癌HCT116细胞增殖、凋亡的影响及机制。方法:用薯蓣皂苷元处理结直肠癌HCT116细胞,CCK-8法测定细胞增殖能力,PI单染法检测细胞周期,Annexin V-FITC/PI双染法检测细胞凋亡,Western blot检测EZH2、C-caspase-3、Cyclin D1蛋白水平。在结直肠癌HCT116细胞中转染EZH2过表达载体,然后给予薯蓣皂苷元处理,同样方法测定细胞增殖、周期、凋亡以及EZH2、C-caspase-3、Cyclin D1蛋白表达。结果:薯蓣皂苷元处理后的结直肠癌HCT116细胞增殖能力下降,细胞G0/G1期增多,凋亡率升高,EZH2蛋白水平下降,Cyclin D1蛋白表达减少,C-caspase-3蛋白表达增多。EZH2过表达载体可以提高薯蓣皂苷元条件下结直肠癌HCT116细胞中EZH2蛋白表达水平,促进细胞增殖并减少细胞G0/G1期阻滞,抑制细胞凋亡。结论:薯蓣皂苷元可能通过下调EZH2抑制结直肠癌HCT116细胞增殖,并诱导细胞凋亡发生。 Objective:To study the effect and mechanism of diosgenin on proliferation and apoptosis of colorectal cancer HCT116 cells.Methods:HCT116 cells were treated with diosgenin,cell proliferation was measured by CCK-8 method,PI monochrome detection cycle,Annexin V-FITC/PI double staining was used to detect apoptosis,the protein levels of EZH2,C-caspase-3 and Cyclin D1 were detected by Western blot.EZH2 overexpression vector was transfected into HCT116 cells of colorectal cancer,then treated with diosgenin,the cell proliferation,cell cycle,apoptosis and the expression of EZH2,C-caspase-3 and Cyclin D1 were also measured by the above methods.Results:The proliferation of colorectal cancer HCT116 cells treated with diosgenin decreased,cell G0/G1 phase increased,the apoptotic rate increased,the expression of Cyclin D1 protein decreased,the expression of C-caspase-3 protein increased,the level of EZH2 protein in HCT116 cells treated with diosgenin decreased.EZH2 overexpression vector can increase the expression of EZH2 protein in colorectal cancer HCT116 cells under diosgenin condition,promote cell proliferation and reduce cell G0/G1 arrest,inhibiting apoptosis.Conclusion:Diosgenin inhibits HCT116 cells and induces apoptosis by down-regulating EZH2.
