骨质疏松性椎体骨折的差异表达miRNA分析及miRNA-mRNA调控网络构建 被引量:1

Analysis of Differentially Expressed miRNAs and Construction of miRNA-mRNA Regulatory Network in Osteoporotic Vertebral Fractures

摘要 目的:筛选骨质疏松性椎体骨折中差异表达miRNA,并构建骨质疏松性椎体骨折中miRNA-mRNA调控网络。方法:通过GEO在线数据库查找骨质疏松性椎体骨折的miRNA表达谱,借助GE02R进行差异表达miRNA分析并筛选差异表达miRNA(|logFC|>1且adj.P值<0.05),借助火山图对差异表达miRNA进行展示,利用TargetScan和miRDB在线数据库预测靶基因,最后通过Cytoscape软件进行miRNA-mRNA调控网络可视化。结果:通过筛选,本研究得到181个差异表达miRNA,其中表达上调98个,表达下调83个。差异表达上调前10位的miRNA包括hsa-miR-205-5p、hsa-miR-660-5p、hsa-miR-155-5p等。差异表达下调的前10的miRNA包括hsa-miR-4508、hsa-miR-1273a、hsa-miR-1253等,其中排名前三的miRNA分别为hsa-miR-205-5p、hsa-miR-660-5p、hsa-miR-155-5p。通过在线数据库TargetScan、miRDB对目标miRNA进行靶基因预测,本研究得出hsa-miR-155-5p预测到280个靶基因,hsa-miR-205-5p预测到304个靶基因,hsa-miR-660-5p预测到294个靶基因。结论:基于GEO数据库挖掘,本研究发现了差异表达miRNA,成功构建骨质疏松性椎体骨折的miRNA-mRNA调控网络图,为骨质疏松性椎体骨折的诊断和治疗筛选出重要的生物标志物。 Objective:To screen the differentially expressed miRNAs in osteoporotic vertebral fractures and construct a miRNA-mRNA regulatory network in osteoporotic vertebral fractures.Methods:In this study,we searched the miRNA expression profile of osteoporotic vertebral fractures through GEO online database,analyzed differentially expressed miRNAs with GEO2 R,and screened differentially expressed miRNAs(|logFC|>1 and adj.P value<0.05),with the help of volcano maps Display differentially expressed miRNAs,use TargetScan and miRDB online databases to predict target genes,and finally visualize the miRNA-mRNA regulatory network through Cytoscape software.Results:Through screening,181 differentially expressed miRNAs were obtained in this study.There are 98 miRNAs with up-regulated expression and 83 miRNAs with down-regulated expression.The top 10 miRNAs with differentially up-regulated expression include hsa-miR-205-5 p,hsa-miR-660-5 p,hsa-miR-155-5 p and so on.The top 10 miRNAs with differential expression down-regulation include hsa-miR-4508,hsa-miR-1273 a,hsa-miR-1253,etc.The top three miRNAs are hsa-miR-205-5 p,hsa-miR-660-5 p,hsa-miR-15 5-5 p.Target genes are predicted for target miRNAs through online databases TargetScan and miRDB.This study shows that hsa-miR-155-5 p predicts 280 target genes,hsa-miR-205-5 p predicts 304 target genes,and hsa-miR-660-5 p predicted 294 target genes.Conclusion:Based on the GEO database mining,this study found differentially expressed miRNAs,successfully constructed a miRNA-mRNA regulatory network diagram of osteoporotic vertebral fractures,and screened important biomarkers for the diagnosis and treatment of osteoporotic vertebral fractures.
