复合酶协同超高压法提取黑木耳多糖的工艺优化 被引量:6

Extraction Optimization of Auricularia auricular Polysaccharides by A Compound Enzyme-Assisted Ultra-High Pressure Method

摘要 该试验研究复合酶协同超高压法提取黑木耳多糖最佳工艺条件。以黑木耳多糖得率为指标,采用单因素试验和正交试验,确定最佳提取工艺参数。结果表明,复合酶提取最佳工艺参数为酶解时间50 min,复合酶(纤维素酶∶木瓜蛋白酶=1∶1,质量比)添加量3%,酶解温度50℃,酶解pH值6.5。在此条件下,黑木耳多糖得率为9.26%。经复合酶法优化后,再进行超高压法提取,最佳工艺参数为保压时间8 min,提取温度50℃,压力400 MPa,料液比1∶30(g/mL)。在此条件下,黑木耳多糖得率为12.23%。 In this study,the optimum conditions for the extraction of polysaccharides from Auricularia auricular by compound enzyme-assisted ultra-high pressure were studied.The yield of A.auricular polysaccharides was used as the index,and the optimal process parameters were determined by single-factor and orthogonal tests.The optimum process parameters for compound enzyme extraction were found to be enzymolysis time of 50 min,compound enzyme addition(1∶1 cellulase:papain,mass ratio)at 3%,enzymolysis temperature of 50℃,and enzymolysis pH of 6.5.Under these conditions,the yield of A.auricular polysaccharide was 9.26%.Ultra-high pressure extraction was carried out after the optimization of the compound enzyme method.The optimum process parameters for ultra-high pressure extraction were holding time of 8 min,extraction temperature of 50℃,pressure of 400 MPa,and solid∶liquid ratio of 1∶30(g/mL).Under these conditions,the yield of A.auricular polysaccharides was 12.23%.
