摘要
为了筛选出具有良好抑菌活性及耐酸、耐胆盐特性的鹅源乳酸菌,试验采用传统的细菌培养方法从健康狮白鹅盲肠中分离细菌,并结合革兰氏染色、生化鉴定和16S rRNA测序方法筛选乳酸菌并对分离菌的耐酸、耐胆盐、耐消化酶能力进行测定,对有高度耐受性的乳酸菌进行抑菌能力、生长特性、产酸能力及生物安全性研究。结果表明:从鹅盲肠内共分离出44株乳酸菌,包括唾液乳酸杆菌19株、罗伊氏乳杆菌和鼠李糖乳杆菌各7株、英格兰乳杆菌和短乳杆菌各4株、约翰乳杆菌3株;从44株乳酸菌中筛选出4株产酸能力较好的乳酸菌,包括2株唾液乳酸杆菌(分别命名为S5、S6)、1株鼠李糖乳杆菌(命名为S8)和1株罗伊氏乳杆菌(命名为S12),这4株乳酸菌能够耐受pH值2~3、胆盐浓度0.2%~0.6%及胰蛋白酶浓度0.8%~1.2%的环境;S5、S6、S8和S12对致病性金黄色葡萄球菌、大肠杆菌、沙门氏杆菌及鸭疫里默氏杆菌均有一定抑制作用,同时具有良好的生长特性、产酸能力及生物安全性,无毒副作用。说明筛选出的S5、S6、S8和S12这4株乳酸菌具有良好的益生菌特性,可用于后续开发鹅用微生态制剂。
In order to screen the goose-derived lactic acid bacteria with good antibacterial activity, acid and choline resistance, the experiment used traditional bacterial culture methods to isolate bacteria from the cecum of healthy Lion-White goose, and combined Gram-staining, biochemical identification and 16 S rRNA sequencing to screen lactic acid bacteria. The acid resistance, choline resistance, and digestive enzyme resistance of lactic acid bacteria were measured. The antibacterial ability, growth characteristics, acid production ability and biological safety of highly resistant lactic acid bacteria were studied. The results showed that 44 strains of lactic acid bacteria were isolated from the cecum of goose, including 19 strains of Lactobacillus salivarius, seven strains of Lactobacillus reuteri, seven strains of Lactobacillus murinus, four strains of Lactobacillus ingluviei, four strains of Lactobacillus brevis, three strains of Lactobacillus johnsoni. four strains of lactic acid bacteria with higher tolerance were selected from 44 strains of lactic acid bacteria, including two strains Lactobacillus salivarius(named S5 and S6 respectively), one strain of Lactobacillus rhamnosus(named S8) and one strain of Lactobacillus reuteri(named S12);these four strains of lactic acid bacteria could tolerate pH values of 2-3, Bile salt concentration of 0.2%-0.6% and trypsin concentration of 0.8%-1.2%;S5, S6, S8 and S12 had a certain inhibitory effect on pathogenic Staphylococcus aureus, Escherichia coli, Salmonella and Riemerella anatipestifer, which had good growth characteristics, acid production capacity and biological safety, no toxic side effect on mice. The results suggested that the four strains of lactic acid bacteria selected, S5, S6, S8 and S12, had good probiotic characteristics and could be used for the subsequent development of microecological preparation for goose.
