太行鸡MC1R基因的克隆测序、表达及生物信息学分析 被引量:2

摘要 为研究影响太行鸡不同羽色性状的重要候选基因MC1R的表达及生物信息学特性,试验以太行鸡麻、白两种羽色为研究对象,采用PCR克隆、实时荧光定量PCR技术和生物信息学分析进行探讨。测序结果显示:太行鸡MC1R基因CDS序列为945 bp,编码蛋白为314个氨基酸;蛋白理化性质分析发现,MC1R蛋白存在7个跨膜区,为跨膜受体;CDS区序列比对发现,太行鸡MC1R基因存在c.637C>T的突变,突变造成编码氨基酸mRNA二级结构和编码蛋白空间结构发生改变;实时荧光定量PCR检测发现,太行鸡麻羽品系毛囊中MC1R的表达量极显著高于白羽品系(P<0.01)。研究结果表明:MC1R基因的c.637C>T突变显著影响太行鸡的羽色,可以作为太行鸡羽色选育的候选分子标记。 The purpose of this study was to investigate the expression,cloning,sequencing and bioinformatics characteristics of MC1R gene in Taihang chickens.PCR cloning,real-time fluorescence quantitative PCR and bioinformatics were used to analyze the relationship between hemp feather Taihang chicken and white feather Taihang chicken.The sequencing results showed that the CDS sequence of the Taihang chicken MC1R gene was 945 bp,encoding protein included 314 amino acids.The analysis of protein chemical properties showed that MC1R protein was a transmembrane receptor protein with seven transmembrane regions.Sequence alignment of CDS region showed that there was a c.637C>T mutation in Taihang chicken MC1R gene.Mutation results in changes in the coding amino acid,the secondary structure of mRNA and the spatial structure of the encoded protein.qPCR detection showed that the expression of MC1R gene in the hair follicle of hemp feather Taihang chicken was significantly higher than that in white feather Taihang chicken.The results showed that the c.637C>T of MC1R gene significantly affected the feather color of Taihang chicken.It could be used as a candidate molecular marker for feather color breeding of Taihang chicken.
