烟酰胺腺嘌呤二核苷酸对急性脊髓缺血再灌注损伤的保护作用

摘要 目的探讨烟酰胺腺嘌呤二核苷酸(NAD+)对脊髓缺血再灌注损伤(SCII)的保护作用。方法将24只大鼠按随机数字表法分为A组(不夹闭主动脉)、B组(主动脉夹闭后注射生理盐水)和C组(主动脉夹闭后注射NAD+)。采用大鼠脊髓损伤(BBB)评分评价神经功能情况,尼氏染色了解脊髓前角正常运动神经元数目情况,测量脊髓组织中超氧化物歧化酶(SOD)活性与丙二醛(MDA)含量来分析氧化应激情况,蛋白质免疫印迹测试脊髓中细胞凋亡情况。结果术后3、6、12、24、48 h BBB评分:B组和C组均低于A组(P<0.05),C组均高于B组(P<0.05)。尼氏染色显示一侧脊髓前角正常运动神经元数目:A组明显多于B组和C组(P<0.05),C组明显多于B组(P<0.05)。术后48 h SOD活性:B组和C组均低于A组(P<0.05),B组低于C组(P<0.05)。术后48 h MDA含量:B组和C组均高于A组(P<0.05),B组高于C组(P<0.05)。Cleaved Caspase-3蛋白表达水平:B组和C组均高于A组(P<0.05),B组高于C组(P<0.05)。结论NAD+通过减弱SCII后的氧化应激反应,保留脊髓前角的运动神经元,对脊髓缺血再灌注损伤起到神经保护作用。 Objective To explore the protective role of nicotinamide adenine dinucleotide(NAD+)on acute spinal cord ischemia-reperfusion injury(SCII).Methods Twenty-four rats were randomly divided into group A(without aortic clamping),group B(injecting saline after aortic clamping)and group C(injecting NAD+after aortic clamping).Basso-Beattie-Bresnahan(BBB)score of the rats spinal cord injury was used to assess the nerve function.Nissl staining was used to understand the situation of normal motor neurons number in the anterior horn of the spinal cord.Measurement of the content of superoxide dismutase(SOD)activity and malondialdehyde(MDA)in the spinal cord tissue was used to analyze the oxidative stress reaction.Western blot was applied to test the cells apoptosis of spinal cord.Results BBB score at 3,6,12,24,48 h postoperation:group B and C were both lower than group A(P<0.05),and group C was all higher than group B(P<0.05).Nissl staining result showed the number of nomral motor neurons in the anterior horn of the spinal cord in one side,group A was significantly more than group B,C(P<0.05),and group C was significantly more than group B(P<0.05).At 48 h postoperation,SOD activity:group B,C were both lower than group A,and group B was lower than group C(P<0.05),the content of MDA:group B,C were both higher than that in the Group A,and group B was higher than group C(P<0.05).The expression level of Cleaved Caspase-3 protein:group B,C were both higher than that in group A,and group B was higher than group C(P<0.05).Conclusions NAD+plays neuroprotective role in spinal cord ischemia-reperfusion injury by reducing the oxidative stress reaction after SCII and preserving the motor neurons in the anterior horn of the spinal cord.
