胶质瘤细胞体外培养技术及应用研究进展

摘要 神经胶质瘤是颅内肿瘤中最常见的恶性肿瘤,因其侵袭程度高,目前治疗效果极差。其具有高度异质性,在肿瘤内部的不同细胞以及肿瘤微环境之间具有复杂的相互作用,因此,我们需要找到一个最佳的培养条件,保持分子基因型和表型以及原始肿瘤在体外和体内的异质性。细胞培养是细胞学研究的基本方法之一,因其独特的优势在各种肿瘤疾病研究中应用广泛。胶质瘤细胞培养方法已基本成熟,主要通过二维(two-dimensional,2D)细胞培养技术和三维(three-dimensional,3D)细胞培养。胶质瘤细胞体外培养主要用于研究胶质瘤的发病机制,同时在药物治疗、放射治疗、免疫治疗等角度为临床工作提供新的治疗思路。本文就胶质瘤细胞体外培养技术及细胞培养的应用进展作一综述。 Gliomas are the most common malignant brain tumors,with poor therapeutic effects due to its highly heterogeneous,which have a complex interaction among different cells within the tumor as well as the tumor microenvironment.Therefore,it is necessary to find an optimum culture condition that maintains the molecular genotype and phenotype as well as heterogeneity of the original tumor in vitro and in vivo.Cell culture is one of the basic methods in the field of cytologic research,it has been widely used in the study of various tumor diseases due to its unique advantages.The glioma cell culture method has been mature,mainly through two-dimensional cell culture and three-dimensional cell culture.The in vitro culture of glioma cells is mainly used to study the pathogenesis of glioma,at the same time,provide new treatment ideas for clinical work from the aspects of drug therapy,radiotherapy,and immunotherapy.This article reviews the in vitro culture technique of glioma cells and its application progress.
