Clearing and fluorescent detection of mice brain by Passive-CLARITY approach

摘要 目的通过Passive-CLARITY法透明化脑组织,并考察透明化过程中脑内荧光信号的变化。方法小鼠经尾静脉注射荧光探针,通过心脏灌流处死小鼠并剖出鼠脑;鼠脑经水凝胶包埋、被动扩散清除脂质;通过成像系统记录脑透明化过程及透明化脑中荧光信号的变化。结果脑组织逐渐透明且体积膨大,在27 d完全透明;透明化过程中,荧光信号逐渐增强,5 d时达到最大值且保持不变。结论采用Passive-CLARITY法可有效透明化脑组织,且荧光探针不随透明化而被清除。 OBJECTIVE To verify brain clearing through Passive-CLARITY approach, and to investigate brain fluorescence during the process of Passive-CLARITY.METHODS Kunming mice were intravenously injected with a fluorescence probe via tail vein, and were then sacrificed by cardiac perfusion.Brains were obtained and embedded in hydrogel solution and were cleared by passive diffusion.Brain fluorescence were recorded via an in vivo fluorescence imaging system.RESULTS Gradual transparency and inflation of brain was observed with time and transparence around 27 d.Brain fluorescence increased during clearing process, and fluorescence signals reached steady state from 5 d.CONCLUSION Passive-CLARITY approach is effective for the clearing of brain tissue without obvious washout of fluorescence probe.
